Paris polyphylla Smith var. yunnanensis (Franch.) Hand.-Mazz. is a variety ofParis polyphylla Smith,which belongs to the family Trilliaceae Paris L.. P.polyphylla var. yunnanensis is a kind of perennial herb. Its rhizome was used astraditional medicine,which is a kind of precious herb. It has been used for thousandsof years in China. Both of P. polyphylla var. yunnanensis and P. polyplylla var.chinensis (Franch.) Hara. were recorded in Pharmacopoeia of People’s Republic ofChina as two biological resource of P. polyphylla. Except P. polyplylla var. chinensis.,another related species were used as P.polyphylla var. yunnanensis, such as Parisvietnamensis(Takht.) H. Li and so on. In this paper, the research of molecularidentification and quality evaluation of P. polyphylla var. yunnanensis andrelated species will be studied via phyto morphyology methods, molecular biologicalmethods and chemical analysis methods.Then help to provide a basis about how tochoose the good germplasm resources of P. polyphylla var. yunnanensis. The mainstudy result is as follows:1. ITS sequence can make a molecular identification among these three cultivatedpopulations: Wuding, Wenshan and Lijiang population. After compared the ITSsequence of cultivated samples and wild samples, we speculated that thegermplasm of these three cultivated populations above-mentioned were P. polyphylla var. yunnanensis more likely; The NJ tree and MP tree based on ITSsequence showed that P. polyphylla var. yunnanensis in the cultivated populationswere transplanted from nearby regions or other cultivated bases very likely.Meanwhile the molecular identification based on ITS sequence also candistinguish Paris vietnamensis from its substitution P. polyphylla var.yunnanensis easily. The CpDNA matK sequence was not fit to make molecularidentification and genetic relationship about the different populations of P.polyphylla var. yunanensis as well as its related species.2. According to the HPLC analysis, The average content of PolyphyllinⅠ,Ⅱ,Ⅵ,Ⅶin the wild populations(3.555%) was2.3times greater than the cultivatedpopulations’s average content(1.570%). P. polyphylla var. yunnanensis is the rawmaterial of Gongxuening Capsule, in condition of ensuring clinical effect andsaving the raw material, it’s better to choose the wild P. polyphylla var.yunnanensis to get the higher content of Polyphyllins; The4Polyphyllins contentin these three cultivated populations detected in HPLC were all higher than0.6%ruled by the Chinese Pharmacopoeia(2010), which showed these three cultivatedbases were meeting the requirement and also had a good quality. We also foundthat Xinping was not the traditional Geo-authentic habitats, but the4Polyphyllinscontent(2.450%) in wild samples were higher.The result suggested us Xinpingcould be the new source of good P. polyphylla var. yunnanensis. The result of NJtree showed that the Wuding wild population with the highest content(6.178%) in4Polyphyllins got together with the Wuding cultivated population with lowestcontent(1.153%) in a cluster, which suggested the content of Polyphyllins in P.polyphylla var. yunnanensis might have no correlation with genotype, and thequality of P. polyphylla var. yunnanensis might be mainly determined by thegrowth environment.3. To compare with P. polyphylla var. yunnanensis according to the HPLC, thecontent of PolyphyllinⅡand PolyphyllinⅦ in Paris vietnamensis was lower thanP. polyphylla var. yunnanensis. Polyphyllin Ⅱ has antitumor effect and immune regulation effect. The result of lower in PolyphyllinⅡalso suggested usParis vietnamensis should not be the substitute of P. polyphylla var. yunnanensis.
