The quality of wheat processing mainly relies on the wheat contained protein.Wheatflour yield also determine whetheran important factor inquality.In this study, wheatspecies “strong gluten wheat Zheng366(A1)” and “weak gluten wheat Zheng004(A2)” were used as materials and the RNA-Seq technology was employed toconstruct the best quality wheat seed transcriptome. And then on the basis of unigenegot from the transctiptome, Bioinformatics methods were employed to mine thedifferentially expressed genes between these two wheat species. The results are asfollows.1. Best quality wheat seed development transcriptome sequencing and the databaseestablishmentWith the high-throughout sequencing technique,63,258,020Clean reads havebeen gained from A1with the cycleQ20ratio of92.35%, while63,258,020Cleanreads have been gained from A2with the cycleQ20ratio of95.12%. After theconting assembling, Scaffolds splicing and Fill hole processing of Clean reads,114,418Unigenes were produced.2. Best quality wheat seed development transcriptome function annotation andmetabolism pathway analysisDatabases like NR, COG, GO, Swiss-Prot and KEGG were used to make thealignment.(1) Nr annotation: the number of unigenes whose CDS area and the sequencedirection can be confirmed is68.787(60.12%in total).(2) GO function analysis:45,708unigenes were annotated with63different kindsof functions, which illustrated that a great deal of genes participate in theprocess of wheat growth and development. (3) GO function classification:13,233unigenes were annotated into25differentkinds of functions, which revealed that genes involved in the protein andstarch synthesis play important roles in wheat seed growth and development.(4) KEGG metabolism analysis:8,629genes have been annotated into118signalpathways among which the ones controlling the protein synthesis andprocessing, hormone regulation, starch and sucrose metabolism provesignificant to the wheat growth and development.3. Best quality wheat related genes miningWith the employment of bioinformatics techniques,375differentially expressedgenes between the Zheng366and the Zheng004were found out. Interestingly,9alcohol soluble protein related genes have significantly higher expression levelin A2than in A1. Besides,11of the375genes have been found participating incarbohydrate transportation and21sorts of enzymes metabolism. Specifically,most of them are involved in the metabolism of glucose-1-phosphateadenylyltransferase、AGPase、1,4-alpha-glucan branching enzyme.This study has provided best quality genes for the wheat processing, which provessignificant to the national wheat food processing and improving the competition ofthe wheat industry.
