In order to elucidate the affect of different soil conditions on characteristics of tobacco style, theflue-cured tobacco cultivar K326growing in dryland purple soil (,Jiangxi, Fuzhou, Longbridge) and sand loampsddy field (Jiangxi, Fuzhou, Ganzhu). The tobacco leaves were managed according to the local high-qualitymanagement measures of production of flue-cured tobacco; In the process of tobacco leaf development,differentstages of middle leaves(leaf10-12) after topping were compared and analyzed on ultrastructure of tobacco leaf,gene expression level on related enzyme gene of plamochromic pigment, main chemical composition,totalfragrant substance of modulation and tobacco smoking assessment. Comparison of different soil conditions affectthe metabolism of tobacco, and metabolism of tobacco style. The main results were listed as fellows:1Ultrastructure:Application of Electron microscopic method to k326of Ganzhu and Changqiao on differentdevelopment organizational structure of the tobacco chloroplast degradation of development, and othercharacteristics, and compares the change of the contents of the cell research. The results show that, in differentsoil conditions, the change tendency of ultrastructure of tobacco leaves on K326were same, relatively fewaccumulate starch grains, small volume changes, more thylakoid lamella content, osmium particles are moreabundant. As leaves mature starch particles accumulate, chloroplast degradation, thylakoid layer disappears, thedisintegration of the nucleus, with osmium particles increases, the cells showed Characteristics of Agingdegradation.Compared to the two kinds of soil conditions, when harvesting osmium particles accumulation growricher. Changqiao in the latter stage of high levels of plamochromic pigment degradation, degradation of starchbetter, faster development.2Use of Flow Analyzer technology analysis of routine chemical components of k326grown in Ganzhu andChangqiao. Discover that (1) The two areas total sugar and reducing sugar contents are shown after the first dropin on an upward trend. Compare different soils, topping the early phase, total sugar content of the Changqiaopurple soil slightly lower, topping30d start, total sugar content in Changqiao is more than Ganzhu, aftermodulation show the same tend.(2) In front of the modulation, starch content of Changqiao is more than Ganzhu,and after the modulation of Ganzhu above Changqiao.(3) The content of nicotine on the rise. Both thedevelopment process and after modulation, the nicotine of Ganzhu significantly higher than the Changqiao, andtotal nitrogen contents were shown a downward trend. The nicotine content of Ganzhu at different times higherthan the Changqiao, after modulation Ganzhu slightly higher than the Changqiao.(4) As the leaf development, potassium contents in a downward trend, after topping30d, the increase of potassium content in two areas aresimilar. After modulation, Ganzhu significantly lower levels of Changqiao. After topping, chlorine has shown adownward trend, longbridge had a slight rebound at later stage, the overall performance is Changqiao higher thanGanzhu. After modulation, the chlorine content of two areas are little different.3Using LC/MS technology test and analyse pigments of k326grown in Ganzhu and Changqiao. Found inaddition to violaxanthin, chlorophyll A, chlorophyll B and lutein and beta-carotene basically the same trend. Thepigment contents of Ganzhu in the various periods are higher than the Changqiao. Especially after topping20dand40d, and after topping30d differences are relatively small. Among them, the promotion of Changqiao as thedevelopment process, pigment content is a smooth downward trend, the pigments of Ganzhu decreased aftertopping10d-30d, after topping30d-40d renders an upward trend. Application of RT-PCR to analysistobacco-related gene expression and found that the chlorophyll biosynthesis-related gene CAB,CHLI have littledifference in the two areas, but chlorophyll degradation expression of related genes PAO, CLH in Changqiao isrelatively strong, especially late-mature leaf development during the harvest. That’s consistent and cellultrastructure observation on the results. Synthesis of carotenoids-related gene PSY, PDS, ZDS, Lcyb, ZEP,Changqiao are higer than Ganzhu. However, the trends of different kind of carotenoid metabolism genes aredifferent in two area.4Neutral aroma and smoking assessment results: Total fragrant substance of roasted Changqiao tobaccowas higher than Ganzhu tobacco, while cembrane degradation product, phenylalanine degradation andcarotenoid degradation products of Ganzhu tobacco were higher than that of Changqiao, and,browning reactionproducts, Changqiao tobacco were higher. Neophytadiene content of Changqiao tobacco was significantly higher.Results of tobacco smoking assessment of Ganzhu product was better,with a total score of27.8, and loweraroma.
