To our knowledge,chicken mitochondrial inheritance is generally assumed to bematernal.Using PCR-RFLP,Hou Lingling invested heteroplasmy in sites mt.ND14589A>G、 mt.ND25703A>T and mt.ND25727T>G in Gushi resourcepopuLation,that indicated mitochondria heteroplasmic universality in chicken. What’sthe relationship between parents and F1generation in heteroplasmictransmission?Hence,we establishment heteroplasmic group,using Pyrosequencingdetect heteroplasmy in sites mt.ND25736T>C、mt.ND25727T>G、mt.ND25703A>T, study the fuLl sibs of chicken F1,estimate genetic model of chicken F1withinND2gene.In order to study the heteroplasmic transmission,we depends on3sites analyzefrom4aspects: neither male parent nor female parent is heteroplasmic; female parentis heteroplasmic while male parent isn’t heteroplasmic; male parent is heteroplasmicwhile female parent isn’t heteroplasmic; both parents are heteroplasmic.The resuLtsshowed that neither male parent nor female parent is heteroplasmic, F1generationheteroplasmic rate in sites5736T>C、5727T>G、5703A>T are5.88%、0%、2.94%,respectively. Correspondingly, female parent is heteroplasmic while maleparent isn’t heteroplasmic, F1generation heteroplasmic rate in sites5736T>C、5727T>G、5703A>T are19.70%、0%、1.52%. Since site5736T>C doesn’t match thecondition of male parent is heteroplasmic while female parent isn’t heteroplasmic andboth parents are heteroplasmic, so we only statistics F1generation heteroplasmic insites5727T>G、5703A>T in the remain conditions. However,male parent isheteroplasmic while female parent isn’t heteroplasmic, F1generation heteroplasmicrate in sites5727T>G、5703A>T are10%、0%,while both parents are heteroplasmic,F1generation heteroplasmic rate in sites5727T>G、5703A>T are4.55%、9.09%.In4aspects,we found F1generation heteroplasmic rate in site5727T>G is0%、0%、10%、4.55%,and in site5703A>T is2.94%、1.52%、0%、9.09%,correspondingly.Meanwhile,we research F1generation genetic model uponheteroplasmic.we respectively detected1individual in sites5736T>C、5727T>G inS(Silkies)♂×S(Silkies)♀and S(Silkies)♂×GA(Rhode Island Reds)♀in94F1generation individuals. We estimated that the rate of paternal leakage was1.06%,which indicated heteroplasmic group was maternal inheritance. This is the firstreport of paternal leakage in chicken. In conclusion, Silkies and Rhode Island Reds are heteroplasmic in3sites inparent generation,and the former is significantly lower than the latter.In F1generation,GA(Rhode Island Reds)♂×S(Silkies)♀and GA(Rhode Island Reds)♂×GA(RhodeIsland Reds)♀in sites5736T>C、5703A>T are heteroplasmic,while S(Silkies)♂×S(Silkies)♀and S(Silkies)♂×GA(Rhode Island Reds)♀in all sites areheteroplasmic. Nevertheless, F1generation heteroplasmic rates are difference in sites5727T>G、5703A>T in4aspects,so is site5736T>C in neither male parent norfemale parent is heteroplasmic、female parent is heteroplasmic while male parent isn’theteroplasmic.What’s more,we did find potential paternal leakage samples in F1generation.
