Polyporus umbellatus is traditional fungus as a kind of food and medicine in china. This test of Polyporus umbellatus cultivation and The test of the Polyporus umbellatus cultivation and filtered the polysaccharide are extracted with hot water extraction optimization, study of the fermentation and polysaccharide solution of several target bacteria and biological activities of plant seeds, in order to provide theoretical basis for the further development and utilization of Polyporus umbellatus. The results are as follows:I.The study of Polyporus umbellatus mycelial growth and polysaccharide content of different culture media has showed that:when we use glucose as the C source and yeast extract medium as N source, the hyphae grows faster, and contains the most abundant polysaccharide; the best Polyporus C N sources for the hyphae growth are glucose and yeast extract; and the optimum liquid culture medium group Polyporus umbellata growth can be divided into:(glucose20g, yeast10g, KH2PO41.0g, MgSO4,7H2O1.0g, VB10.lg). Under this culture condition, the dry weight of mycelium in each bottle is2.65g, dry mycelium and fermentation broth polysaccharide content being respectively13.25mg/g,6.63mg/100mL.Ⅱ.Study on the effect of different factors on the hyphae of Polyporus umbellata polysaccharide content has showed that:the optimum particle size cultured mycelium of Polyporus umbellatus liquid is200; the leaching bacteria powder and solvent volume ratio of is1:30, with extraction time being2.5h, extraction temperature85℃, pH7, extraction3times; ratio on the yield of polysaccharides has no effect; ethanol precipitation agent ratio is3:1.Ⅲ. The research of Polyporus umbellata fermented liquid and polysaccharide solution in e. coli, hay bacillus coli, wheat fusarium graminearum, pathogen of tomato leaf mold, made dry silk nuclear bacteria, penicillium six target bacteria activity of has showed that:two kinds of solution on e. coli and hay bacillus coli inhibition, the greater the concentration, the stronger antibacterial activity. Two solution is sensitive to acid and alkali, fermented liquid and pH6pH7bacteriostatic activity of the largest, and the polysaccharide solution are pH5and pH6bacteriostatic activity of the biggest; Fermented liquid strongest antimicrobial activity in40℃,35℃less than or greater than the loss of bacteriostatic activity of45℃, the polysaccharide solution the strongest antimicrobial activity in35℃, the temperature range of30~50℃. Chloroform organic solvent extraction effect is best. Hay bacillus coli conductivity with the extension of treatment time increased, the MDA content increased after the first reduce trends.Ⅳ.The research of Polyporus umbellata fermented liquid and polysaccharide solution of cucumber, watermelon and tomato seed activity has showed that the two kinds of solution have germination inhibition effect on3seed, and the inhibitory effect of Polyporus umbellata is stronger; the conductivity of Cucumber seed increases along with the increment of2kinds of solution concentration, and content of MDA increases with the increase of concentration.
