Ameba trifoliate (Thumb.) Koidz. is a deciduous woody vine, which belongs to Lardizabalaceae, distributed in the Yangtze River basin, south Shaanxi (Hanzhong, Shangluo prefecture majority), Shanxi Jinnan area, Gansu southeast and Chongqing area, and also distributed in Hubei, Hunan, Sichuan, Shandong, Jiangsu, Hebei, Jiangxi, Zhejiang, Henan, etc. The dry cane can be used as traditional Chinese medicine and dry nearly ripe fruit can be used as medicine, called Akebia Fruit. Study proves that the main effective components of Akebia trifoliata are saponins kind material, in addition, fruit, root, seeds and leaves also contained many kinds of chemical compositions such as some flavonoids, grease, amino acids, polysaccharide, phenols, steroid, lignin class and so on. Flavonoids and polysaccharides material research rarely for Akebia trifoliata(Thunb.) Koidz. Flavonoids and polysaccharides compounds have antioxidant, improve human disease-resistant ability, delay ageing, anti-inflammatory cancer and so on.This thesis optimizes extraction of flavonoids of Akebia trifoliata(Thunb.) Koidz by the ultrasonic technology and microwave-assisted extraction technique. Flavonoids contents of Akebia trifoliata(Thunb.) Koidz are determined from different parts, different origin and different age order. For the better development and utilization Akebia trifoliata(Thunb.) Koidz, provides the basis. The cottonwood flavonoids are predicated using macroporous adsorption resin. We compare purification things with crude extractings antioxidant. Akebia polysaccharide monosaccharide is examined by reversed-phase HPLC. We also research the oxidation of polysaccharides and the lead role. The main results of experiment are as follows:1. Flavonoids content in root is highest through the determination, up to5.72%. The flavonoids content is the lowest in pulp, only0.2%. Flavonoids contents of seeds and pulp are no significant differences. Flavonoids contents of leaf,25to27-born stems,8to10year born stem are no significant differences. Akebia cottonwood16years old flavonoids content is the hightest of9.34%, born in9flavonoids content is the lowest of3.17%.8and14years old flavonoids content are no significant differences. Born in12and born in10are no significant differences.18and19years old are no significant differences.2. Flavonoids contents are determined from seven provinces. The date analysis by SPSS17.0found that akebia cottonwood flavonoids contents in different origins have significant differences. Flavonoids contents of seven provinces are in the order of Hunan>Hubei>Guizhou>Shaanxi>Henan>Gansu>Shanxi. Flavonoids contents of sixteen origins are in the order of Cili County, Zhangjiajie in Hunan>Siping village, Hefeng County in Hubei>Shimen County in Hunan> Jiudongtian village,Sangzhili County in Hunan>Shengnongjia songbai in Hubei>Zhenyuan in Guizhou>Shengnongjia muyu village in Hubei> Sanzhi County Hongjiaguan in Hunan> Yinping village Zhenan in shaanxi>Zhenan huinongzhen zhajiagou in Shaanxi> Zhenan Akebia base in Shaanxi> Wuliqiao tiandingdong village, Xixia County in Henan>Zhenan xiaojiagou in Shaanxi>Wafang village, Xixia County in Henan>Liangdang yuchi town in Gansu>Yuanqutianpan mountain in Shanxi. On the basis of methodology of the experimental results, RSD value of precision, stability and the sample recovery are less than3.0%. Experimental system is stable and method is reliable.3. Flavonoids process is studied through the ultrasound auxiliary method and the microwave-assisted extraction method, results as following:(1) Influence factors of ultrasonic auxiliary extraction are in the order:extraction times> liquid materials>extraction time>ethanol concentration. It means extraction times are the biggest influence factor for cottonwood extraction, then material liquid, and ethanol concentration affects the minimum. The optimal conditions:extraction times three times, material liquid (g:mL)1:30, extracting time30min.SPSS17.0statistical software variance analysis shows that extraction times have a significant influence.(2) Influence factors of microwave-assisted extraction are in the order: microwave power>ethanol concentration> liquid materials>extraction time. That power of microwave extraction are the biggest influence, ethanol concentration is the second. The optimal conditions:extraction times three times, material liquid (g:mL)1:30, extracting time30min. SPSS17.0statistical software variance analysis shows that extraction times have a significant influence.4. Through choosing the AB-8, D101, HPD722, HPD826, HPD450and HPD600macroporous adsorption resin, HPD722is finally selected as purification of akebia cottonwood flavonoids resin. Processed HPD722resin fills into the column (40x1.6cm), volume for two-thirds of the column. Resins are washed using100ml distilled water with the speed of1ml/min, then add in the prepared sample liquid225ml in adsorption by1ml/min velocity, finally wash down flavonoids with the60%ethanol100ml,5. Akebia cottonwood coarse and purified flavonoids oxidation resistance are compared through the DPPH free radicals system, hydroxyl radicals (· OH) system and restore ability system. Results showed that coarse flavonoids and purified flavonoids all have certain antioxidant activity, but purified flavonoids oxidation resistance is better than coarse flavonoids. Its antioxidant ability is after BHT. We can consider using akebia cottonwood flavonoids antioxidant effect instead of BHT.6. Simple sugars comprised akebia polysaccharide is detected by HPLC: D-mannose, L-rat lee sugar, D-half of lactose hyaluronic acid, D-glucose, D-half of lactose and L-Arab sugar.7. Akebia cottonwood polysaccharide oxidation resistance are compared through the DPPH free radicals system, hydroxyl radicals (· OH) system and restore ability system. Results showed that polysaccharide have very good antioxidant activity. When they reach a certain concentration, its antioxidant ability is after VC. This study provides the theory basis for the development of non-toxic antioxidants.8. We research akebia polysaccharide eliminating lead for the mice in the test. Results show that it has certain effect of eliminating lead.
