In this experiment, useing Opisthopappus (Opisthopappus taihangensis, Opisthopappus longilobus) and multiple chrysanthemum varieties as materials. By doing fluorescence microscopy pollen adhesion in the process of pollination and Pollen germination (hybridization between Opisthopappus and chrysanthemum) to study how the different hybridized combination, different pollination methods, boric acid, Ga2+, NaCl influence hybridization affinity. By observing the embryo development after pollination of different hybridized combination to decide time of hybrid embryo abortive. To rescue hybrid embryos, we determined the most appropriate embryo culture starting time, the best hybrid combinations and embryo culture medium, provide theoretical basis for distant hybridization between opisthopappus and chrysanthemum.From the experiment, the results showed that:Different concentration of boric acid, Ca2++100mg/L suga for the medium has no significant impact on germination and pollen adhesion on stigma. the hybrid combinations of Opisthopappus taihangensis and’saimudan’ is conducive to pollen adhesion on stigma, and pollen germination are normal;By applying the mixed pollen pollination on the hybrid combinations of Opisthopappus taihangensis and ’saimudanis’ is conducive to the pollen adhesion and germination.After sprinkling different concentration NaCl on stigma, pollen adhesion on stigma and pollen germination are influenced. Stigma was in8%NaCl treatment for0.5h, observation of pollen adhesion the highest number, pollen germination is norma24h after pollination.Different hybrid combinations have appeared the phenomenon of hybrid embryo abortion, a lot of hybrid embryo abortion period generally occurs in early embryonic development,Torpedo-shaped embryos and embryos during the heart,hybrid embryo abortion is an important factor affecting distant hybridization barriers.Hybrids combination of Opisthopappus and Ground-Cover chrysanthemum best embryo rescue period is10-15days after pollination, Opisthopappus and ’dahong’ embryo callus was the best among different hybrid combinations, the best embryo culture medium was MS+BA2.0mg·L-1+NAA0.2mg·L-1.
