Object: Qingfeizhike Granule combine classical antipyretic agent honeysuckleflower-baikal skullcap root and antitussive balloonflower root-ansu apricot seed, togetherwith another two kinds of drugs white mulberry root-bark and common anemarrhena tostrengthen the function of relieving cough and asthma, regulating vital energy. The contentsof these components in Qingfeizhike Granule were determined to investigate their toxicologyand pharmacodynamics. Morever, the clinical effects of Qingfeizhike Granule in the researchcan do help for the clinical application for veterinary.Method:(1) Assay of these components: Adopting the ultraviolet spectrophotometry toevaluat the contents of honeysuckle flower and baikal skullcap root by assaying their majoringredient chlorogenic acid and baicalin respectively.(2) Acute toxicity tests: Acute toxicitytests and maximally tolerated dose test judge the safety of Qingfeizhike Granule.(3)Pharmacodynamics: Xylene-induced auricle inflammation experiment, rabbit pyrogenic test,strong ammonia solution-induced cough test determine how is the function of anti-inflammation, relieving fever and cough.(4) Clinical effects of Qingfeizhike Granule:Treated animals with Qingfeizhike Granule are experimental group and apricot kernel coughpill cured animals are as control. Next, Compare effects of the two group and decide itsclinical effects.Results:(1) Assay of these components: chlorogenic acid shows a good linear relationwithin13.2~66.0μg/mL while within2.5~15.0μg/mL for baicalin. Additionally,coefficient of recovery and precision all fill the bill, suggesting its feasibility. And the contentof chlorogenic acid is2.562μg/mg and that of baicalin is1.4923μg/mg.(2) Acute toxicitytests: Maximally tolerated dose of Qingfeizhike Granule is320g/kg, which suggests that theyhave so low toxicity that satisfy the general toxicity of veterinary drugs requested by Ministryof Agriculture.(3) Pharmacodynamics: Qingfeizhike Granule can elongate the latency periodof strong ammonia solution-induced cough and decrease the cough frequency in2min formice, inhibit rabbit fervescence induced by peptone and auricle inflammation. These testsindicate that Qingfeizhike Granule have strong functions such as anti-inflammation, relieving fever and cough.(4) Clinical effects: The recovery rates to cure three groups of pig coughwith lung heat by high, medium and low dose of Qingfeizhike Granule are85%,70%and30%, and the total effective rates are95%,95%and55%. The result showed QingfeizhikeGranule can be used in pig cough with lung heat without side effect.Conclusion: The content of chlorogenic acid in Qingfeizhike Granule is2.562μg/mgand that of baicalin is1.4923μg/mg, the result showed the ultraviolet spectrophotometrymethod is simple, quick and reproducible. It is a kind of safe Chinese drugs pharmaceuticswith low toxity. Pharmacodynamics shows its functions are anti-inflammation, relievingfever and cough. Clinical effects experiment indicates it can be used in curing pig cough withlung heat without side effect.
