The seed purity and genuineness are key factors to ensure potential varietyincrease and product output. Conventional seed identification technique is timeconsuming and is subject to environmental or time conditions, which is difficult tomeet market needs. Development of rapid and accurate identification technology inseed purity and genuineness is benefit to protect the interests of farmers, to save timeand grasp business opportunities for seed production unit. Study on the mainadvantage vegetable in Tianjin city of Chinese cabbage and cauliflower, closelyaround the key point of "seed purity and authenticity" in production of vegetablevarieties, based on research of previous work, available resources EST of vegetablevarieties, comprehensive EST-SSR and EST-SNP new generation of molecularmarkers, screened primers complementary between the parents and hybrids,construction EST-SSR and EST-SNP markers fingerprinting of Tianjin productionadvantages of vegetable varieties, and further using the pyrosequencing, andhigh-resolution melting curve analysis by polyacrylamide gel electrophoresis todetected the difference fragments or difference locos, establish a rapid, accurate,high-throughput new seed purity and genuineness detection system. The mainfindings are as follows:1、DNA extraction method from single seed was build with the advantages ofsimple operation, time-saving, economic-saving and good Practicability.2、The seed purity and genuineness of identification primer library of Chinesecabbage was selected from305primer pairs, including30pairs of core featuresprimers;Two cabbage composite EST-SSR fingerprinting were developed12Chinesecabbage can be accurately divided into three categories; Found7EST-SSR markersloci for seed purity identification, identification results consistent with themorphological identification field;A Chinese Cabbage EST-SNP fingerprint and twoSNP locus were founded to differentiate Chinese Cabbage varieties, the two locuscould identify nine varieties purity successfully, and consistent with SSRidentification results..3、The seed purity and genuineness of identification primer library of cauliflowerwas selected, including36pairs of core features primers;A cauliflower compositeEST-SSR fingerprinting were developed19cauliflower can be accurately divided intothree categories; EST-SSR markers for seed identification were found to identify thepurify of four identification results are consistent with the morphologicalidentification field.
