Maize (Zea mays L.) is one of major grain crops in China, seed quality is a major outstanding problem in agricultural production. The identification of genuineness and purity of maize seed quality is an important indicator of seed inspection, to keep the fake and shoddy seeds from the market. The objective of this thesis is to construct the technical system for the detection standard of genuineness and purity of hybrid maize seed and its inbred line through the optimization of standard SSR identification system, selection of core primer and establishing the standards of bands interpretation.The main results were summarized as follow:(1) SSR technique was optimized on genomic DNA extraction, PCR procedure and electrophoresis detection. The analysis method fitting for SSR markers of maize was established based on these options.(2) 127 SSR primer pairs were chosen according to literature and maize GDB databse(http://www.maizegdb.org). 16 maize crossing combinations and 32 main maize inbred lines were analyzed using these SSR primers. Based on the balanced distribution in chromosomes, stability, high value of PIC, reasonable distribution, a set of 40 core primer pairs was selected (consists of 20 pairs of and 20 pairs of class IL).(3) Based on above study, the identification system of genuineness and purity of hybrid maize and its inbred line were studied systematically. And an initial standardized criterion of interpretation were established, the effectiveness and feasibility of the identification system were verified through the detection of artificially mixed populations (3 inbred lines and 3 hybrid lines) by the established system.
