Mallard is one duck ancestor mallard ducks for my country.Once wild duck population in our country is relatively abundant,but due to excessive hunting and habitat conditions change,the population is decreasing.Wild ducks are widely distributed,most of them are migrating birds,it is the important economic birds in China.However,studies show that the virus detection rate of anatidae birds is much higher than other birds,as the migratory waterfowl in the process of the influenza virus spread play an important role.As an important species of migratory waterfowl.Anas platyrhynchos play a certain role in the process of the spread of the virus.Major histocompatibility complex widely exists in vertebrates and code immunoglobulin-like receptors and it is a kind of highly polymorphic gene group.Its production of MHC have much relations to immune system,growth and development,reproductive physiology and aging and determines the compatibility of the organization.often as functional gene molecular markers to research the genetic diversity and to analysis population disease resistance,confirmed that the resistance of the related species in different haploid type of influenza virus are difference?In this study mallard ducks is the research object through c DNA to ensure the expressed loci in exon2 and exon3.to get all of the expressed loci by RACE technology.And depend on the consequence to design the unit point amplification primer(exon2andexon3),due this to establish the unit amplification system.Further more do the PCR-SSCP,polymorphism analysis of MHC class I for 50 mallard,and do classification to do genetic diversity analysis.The results shows that mallard ducks have two MHC class I expressed loci Through the consequence of amino sequence aligentment and cluster analysis by multiple species MHC I genes showed that mallard ducks has the most intimate relationship,followed chicken.Through the group survey that exon2 and exon3 all have high polymorphism and the main method is mutation.The content of GC is 59.6% and 51.2%,the content of GC in the third codon is 89.7% and 73.6%,this shows exon2 under the more pressure of mutation.This related to the function of exon combined with antigen.Group analysis shows that MHC class I genes of exon2 and exon3 far from balance(p<0.05),the PIC He Ne of exon 2are 0.460?3.784?0.68,the PIC He Ne of exon 3 are 0.460,3.211,0.635.The figures show that mallard ducks group with high genetic polymorphism.This study reveals the mallard ducks population genetic diversity of MHC,for the protection of the species and provide certain theoretical basis for the use of genetic breeding.
