| Based on reproducible ewe and advanced science-technology means, modern mutton sheep production system can make full use of ewe’s breeding potential and improve the fertility. While the breeding of Ning-xia Tan sheep is still stay in the traditional natural mating, which leads to low low fecundity. Another reason is that vale of the important means of advanced technology at home and abroad isn’t be played and reflected. At last, reproductive technologies such as synchronization of estrus and artificial insemination is not widely available.According to current situation of Ning-xia Tan sheep’s breeding, we studied on the applicability of synchronization of estrus and semen cryopreservation technology. Among different treatment schemes of synchronization of estrus, the best one could be found. By means of studying on results of different ram semen cryopreservation diluents formulas, we picked out the optimum formula. In order to exploring techniques of improving Tan-sheep’s reproductive capacity, a series of breeding technology such as synchronization of estrus, detection of oestrus, artificial semen collection, semen dilution preservation techniques, artificial insemination and pregnancy diagnosis.etc were combined. This experiment research results are as follows:1. The estrus synchronization rate and conception rate of the group which treated with “NIDR + FSH + PG†is higher than that of group treated with “PG + FSHâ€(P <0.05). the best scheme is: set up NIDR after injection “FSH 50 IU + PG 0.1 mg†96 hours with treating 13 days, within-24 h,-12 h injection FSH 50 IU, 0 h remove NIDR and injectioning PG 0.1 mg the same time, which would lead ewes estrus synchronization rate to 100%, very significantly higher than other groups(P<0.05), and conception rate to 75.3%, significantly higher than other groups(P<0.05), furthermore, estrus time was mainly concentrate in 24-48 h。2. The No.5 presented a good preservation among the six kinds of semen diluents formula collected in this study, the preservation time, effective survival time, survival index were significantly higher than other groups(P<0.05), No.6 comes second. Preservated by No.5 diluent, tan sheep’s sperm surviving for 162 hours longest, effective survival time reached 87±5 h, survival index was 89.64±5.1, and the conception rate above 50% within 24 hours.3. The addition of trehalose to semen diluter can raise sperm viability, sperm motion parameters and integrity of sperm plasma membrane(P<0.05). in the concentration range of this study designed, by trehalose concentration increasing, the cell viability no significant change(P>0.05), the integrity of sperm plasma membrane and motion performance of sperm showed an increasing tendency, but this tendency was not significant(P>0.05), when the concentration is 0.03 g/mL, the integrity of the plasma membrane was higher than that of other concentrations, and the difference was not significant(P>0.05), the sperm velocity was higher than other added concentrations also, the difference was not significant(P>0.05). |
