At present,there are many kinds of injection of compound anesthetic for dogs.However,compound anesthetic exits so many problems, such as poor effect of anesthesia, individual differences, low security, strict supervision of drugs, high price and lack of antagonists. In less-developed regions, injection of compound anesthetic is used in many zoo hospital widely.Thus,the research on compound anesthetic with good effect of anesthesia,high security and low price still has great significance.In this study, dexmedetomidine and midazolam are chosen to the main component of the compound anesthetic.According to the principle of balanced anesthesia, applying preliminary experiments in mice,drug screening test,a combination of orthogonal test and righting reflex,anesthesia score test,biological reflex test and basic physiological indexes for monitoring test to make sure the best ratio of dexmedetomidine and midazolam was 0.25:0.18,and this ratio was called DM.After the administration of DM 0.1mL/kg(IM),all dogs stood unstably、fall on the ground,and then went into anesthesia. Applying anaesthesia scores scale to assess the anesthesia effect of DM,the results showed that sedation,analgesia and muscle relaxation were in good quality.The dogs anesthetized by DM were in short induction,suitable maintenance time and stable recovery.The monitors of basic physiological parameters showed that DM had influence on cardiopulmonary parameters, heart rate and respiration rate decreased by 38% and 34%,and the blood pressure increased firstly,and then decreased to normal until 60 min.Using Datex-Ohmeda S/5 to minotor Pet CO2, TV and MV, and the results showed that PetCO2,TV and MV maintained 36~38 mm Hg,106~112 m L and 1.48~1.69 L,and these were in normal physiological range.These results revealed that DM had few influence on respiratory system. And Pa CO2、Pa O2、pH、HCO3-、BE、Sp O2 were in normal physiological range too.The results of electrocardiogram showed that DM had influence on PR,and it was a little beyond the normal range of dogs. During the whole procedure,Q wave,R wave,S wave and T wave were in the normal range. Besides T wave had a trend to decrease at the beginning of anesthesia,but it turned to normal at the end of anesthesia.During the surgery, all dogs could be compeleted the procedure smoothly, and Sp O2 maintained above 95%. There were no significant difference in HR、RR、SPO2、SBP、DBP、MAP at same time point when compared with control group except T wave(P > 0.05).Inconclusion,DM can provide anesthesia for 40~50 min,and the effect of sedation, analgesia and muscle relaxation are in good quality. Besides, DM has few influence on circulatory system,respiratory system and cardiovascular system,it can be used for dogs.
