The first genetic linkage map of Salvia miltiorrhiza was constructed in 94 F1 individuals from an intraspecific cross by using SSR, SRAP and ISSR markers. The results obtained in this study were as the follows:1 construction of genetic linkage mapA set of 928 SSR primers, 100 SRAP primers and 8 ISSR primers developed from Salvia miltiorrhiza were screened in the two parents. O f them, a total of 471 primers yielded polymorphic amplicons, respect with 423(45.6%) SSR primers, 47(47%) SRAP primers and 1(12.5%) ISSR primer. A total of 150 marker loci in the linkage map, consisting of 102 SSR, 46 SRAP and 2 ISSR markers were made up of eight linkage groups, three linkage pairs, covered a total length of 527.7 c M with an average distance of 3.5 cM per marker. The length of linkage groups varied from 26.4-131.6 cM and each of them included 8-29 markers respectively. It was estimated that the Salvia miltiorrhiza genome genetic distance was 544.8 cM representing 93.5% genome coverage.2 QTLsHereditary variations of 6 root traits were measured and analyzed based on fimapping population of Salvia miltiorrhiza. The results were shown as follows: There were 19 QTLs for these 6 root traits using interval mapping, phenotypic variance explained ranged from 18.3%-28.1%.3 study on transferability of Salvia miltiorrhiza EST-SSR markersIn this paper, 311 pairs of EST-SSR primers, designed from genomes of Salvia miltiorrhiza, were employed to evaluate primer transferability in the genomes of 5 labiatae medicinal plants. The results show that 272 pairs of EST-SSR primers were successfully amplified in the genomic DNA of Salvia miltiorrhiza. The transferability ratios o f EST-SSR primer pairs from S. przewalskii, Scutellaria baicalensis, Perilla frutescens, Mentha haplocalyx, S.splendens were 54.8%(149), 44.5%(121), 31.6%(86), 42.6%(116), and 50.0%(136) respectively with an average of 44.7%. The results indicate that the Salvia miltiorrhiza EST-SSR marker is highly transferable to other labiatae medicinal plants.In summary, this study may facilitate genomic structure and function, maker-assisted selection breeding, and comparative genomic analyses between Salvia miltiorrhiza and relative species, may further provide an important reference for molecular marker breeding of Salvia miltiorrhiza, and pave the way for the study of genetic mechanism of Salvia miltiorrhiza.
