Objective: To investigate the germplasm source, cultivation history, and processing overview of Bo Baizhi. To determine the content of 16 kinds of inorganic elements in 12 samples of Bo Baizhi, and to explore the enrichment regularity of the inorganic elements in Bo Baizhi root and foliage during different harvest time. To evaluate the quality of Bo Baizhi. In addition, to explore the relativity between inorganic elements and the quality of medicine, and to study dynamic changes of two components content during different harvest time. To search for habitat processing method replaces sulfur-fumigation for Bo Baizhi. To establish the HPLC fingerprint of Bo Baizhi.Methods: The spot survey for Radix Angelica dahurica planting in the area of Bozhou. Bo Baizhi root, foliage and the soil near the root are treated by microwave digestion method, and are measured the content of 16 kinds element in the ICP-MS method of KED mode, which the correlation analysis was performed by using SPSS 19 software. Using the method of HPLC measures the content of imperatorin and isoimperatorin, which the mobile phase is methanol-water(65:35), the detection wavelength is 300 nm, the flow rate is 1.0 m L·min-1, and the column temperature is 30℃.The same batch of Bo Baizhi(fresh) is treated by 7 different drying methods, which is measured the content of imperatorin and isoimperatorin with HPLC method. Using the method of HPLC, taking acetonitrile-water as mobile phase with gradient elution, detection wavelength is 300 nm, the flow rate is 1.0m L·min-1, column temperature is 25℃, to establish the fingerprint of Bo Baizhi. Using “similarity of chromatographic fingerprint of traditional Chinese medicine evaluation system”( A version 2004) and SPSS 19.0 software for analysis.Results: Bo Baizhi is mainly introduction from Yuzhou city of Henan province, its medicinal properties most similar to Yu Baizhi. The content of 12 samples of BoBaizhi, in which K、Ca、Mg、Na、Al、Ba elements is relatively high and 4 kinds of heavy metal Cu, As, Cd, Pb elements shall not exceed the standard. In the different harvest period of Bo Baizhi root and foliage enrichment of 16 elements is not the same, where the enrichment of the Mo element is relatively strong in the root and foliage. The average content of 12 samples of imperatorin and isoimperatorin in Bo Baizhi is respectively 0.271%, 0.098%, and there is a significant positive correlation between Al、K、Ca、Cr、Ba elements and two active ingredients. Four different producing areas of two active ingredients average content in Radix Angelica dahurica is Qi Baizhi>Bo Baizhi > Chuan Baizhi > Yu Baizhi. In different harvest time, the two active ingredients content of Bo Baizhi is to reach the maximum value in August. Using seven kinds of drying methods, a maximum total content of the two components after the microwave drying process is 0.592%. To establish the HPLC fingerprint of Bo Baizhi, which non-sulfur-fumigation demarcate 15 common peaks, while sulfur-fumigation demarcate 9 common peaks, and their similarity of spectra is 0.879. There is a high similarity between the four origin of Radix Angelica dahurica.Conclusion: Bo Baizhi species is Angelica dahurica(Fisch.ex Hoffm.) Benth.et Hook.f.. To analyze the correlation between inorganic elements and the quality of medicine, and enrichment regularity, which can provide scientific basis for improving the quality and yield of Bo Baizhi. Compared with traditional commodities Angelica dahurica, the quality of Bo Baizhi is good, so Bozhou can be used as a new primary areas. The Microwave drying method can be used as the process method in the producing area of Bo Baizhi, the specific process parameters of microwave drying has yet to be studied further. The method to establish the fingerprint of the test is simple, with good reproducibility. With establishment the fingerprint of non-sulfur-fumigation and sulfur-fumigation Bo Baizhi, can be used for the qualitative identification.
