| In order to find the local varieties of Anhui province chaohu ducks’population genetic diversity and meat flavor matter inosine monophosphate (IMP) content, as well as the impact of IMP abundance gene polymorphism. This experiment used 14 microsatellites marker in 96 chaohu ducks to analysis the population genetic diversity of chaohu duck, with high performance liquid chromatography (HPLC) method to determine the IMP content in chaohu ducks’ breast muscle and leg muscle. The combination of PCR-SSCP and sequencing method were used to detect the IMP synthesis process which is one of the key enzymes of adenylosuccinate lyase (ADSL) gene polymorphism, and 12 weeks of chaohu ducks’ breast muscle and leg muscle inosinic acid content in correlation analysis. The results are as follows:1. Microsatellite marker in chaohu duck group detected a total of 48 alleles. The average allele number per loci was 3.43. The genetic gene frequency distribution was between 0.01 and 0.97. The polymorphism information of the fourteen microsatellite seats content was between 0.19 and 0.95. The effective number of alleles was between 2 to 6, and the genetic heterozygosity ranged between 0.19 and 0.95. Population genetic diversity showed that the genetic is rich, and it can be used for directional breeding of chaohu duck in the future.2. The male duck breast muscle IMP content was 0.44 ±0.23 mg/g, leg muscle IMP content was 0.36 ±0.24 mg/g, the female duck breast muscle IMP content was 0.57 ±0.27 mg/g, and the leg muscle IMP content was 0.36 ±0.19 mg/g. The male duck IMP content in breast muscle was significantly higher than that of leg muscle (P<0.05), the female duck IMP content in breast muscle was significantly higher than that of leg muscle (P<0.01), breast muscle between male and female IMP was significantly (P<0.01), but the content of leg muscle IMP no significant difference between male and female (P>0.05).3. To amplificate 17908 bp to 18066 bp of ADSL gene, the stripe analysis and sequencing of five genotypes was detected, AA, CC, AB, BC and AC type were respectively named. Genotype frequencies were 0.66,0.17,0.08,0.02,0.07, and the frequencies of the alleles of A, B and C was 0.74,0.05 and 0.21. The heterozygosity of the locus gene was 0.41. The site in a moderate polymorphism (PIC=0.41), chi-square suitability test results show that the proposed site genotype distribution was in Hardy-Weinberg equilibrium (χ2=1.33, P>0.05). AB compared with the other four genotypes base T insertion in bp 17948, compared with other four kinds of genotype CC type base C in 17953 bp insertion and existing C18001T, T18003C, G18010C three point mutations, BC type comparing with the other four genotypes existence T17965C point mutations, AA, CC, compared with the other three genotypes in 18091 bp in the lack of base C.4. To amplificate 18158 bp to 18384 bp of ADSL gene, the stripe analysis and sequencing of three genotypes were detected. DF, EE and FF type were respectively named. Genotype frequencies were 0.05,0.85 and 0.10, respectively, D, E, F frequency of alleles were 0.03,0.85,0.12, The heterozygosity of the locus gene was 0.26, The site in a low polymorphism (PIC=0.23), chi-square suitability test results show that the proposed site genotype distribution deviates from the Hardy-Weinberg equilibrium (χ2=13.22, P<0.01). DF type and has been published in NCBI mallard ducks of ADSL (NW-004678857) identical gene sequences, the wild type, define it as the other two genotypes compared with wild type in the loss of 18216 bp in base A, EE type compared with wild type G18225 A point mutations exist and the fragment in 18259 bp TTTATTGCCTTTC lost A total of 13 bases.5. ADSL gene polymorphism loci of eight genotypes and chaohu duck breast and leg muscle IMP content had no significant correlation. Combined genotype and chaohu duck breast and leg muscle IMP content analysis found that the IMP content differences between AAFF and CCFF, CCDF and CCFF, AAEE CCFF ACEE and ACEE were significantly correlation(P<0.05).As a result, the population genetic diversity of chaohu duck was rich; The male duck breast muscle IMP content was significantly higher than leg muscle, female duck breast muscle IMP content was significantly higher than leg muscle, breast muscle between male and female of IMP content were differences significantly, and leg muscle were no significant difference between male and female IMP content. Chaohu duck ADSL gene insert bases, missing pieces, missing bases and point mutations in a total of four types of mutations in 11 introns. Multiple locus mutation makes significant difference between breast and leg muscle IMP content in chaohu duck. |
