The excellent clones of Larix potaninii Batal (Y1、Y2) in Guangxi were chosen to explore tissue culture and rapid propagation technology. The explant initiation culture conditions and external factors on the proliferation of Plantlets in Larix potaninii Batal were on Study, the seedling rooting culture medium and the kind and concentration of Exogenous Additives were screening. To select the best micro cutting conditions of different Larix potaninii Batal clones. In order to analysis the relationship between physiological indexes and tissue culture effect of different Larix potaninii Batal clones the stage of tissue culture, the main research results are as follows:1. The disinfection of 0.1% mercuric chloride 6 min and the induced medium of MS+6-BA 1.0 mg/L+ IB A 0.3 mg/L is suitable for the Larix potaninii Batal explants. Using 0.1% HgCl2 treatment 6 min, the contamination rate was low, at 35%, and less explant browning degree; Using MS+6-BA1.0mg/L+IBA0.3mg/L as explants induction medium, explants of germination rate was 13.33%. Along with the increase of processing time, the content of total phenol was increased and the activity of PPO was decreased, the browning degree of Y1 clones was more serious than Y2, the processing time was longer,and the more serious in the browning. And the anti browning ability of Y2 was better than Y1.2. The different clone of Larix potaninii Batal, the optimal proliferation medium and the suitable plant growth regulator and concentration was different. Shading was not conducive to the accumulation of chlorophyll, it had little influence on the tissue culture seedlings of browning in tissue culture whether is shading. And the Tissue culture ability of Y2 was better than Y1. The best subculture proliferation medium of Y2 was 6-BA 0.6mg/L, IBA 0.3 mg/L, KT 1 mg/L and 6-BA 0.4 mg/L+IBA 0.1 mg/L+KT lmg/L, and the proliferation rate up to 4.60; The best subculture proliferation medium of Y1 was 6-BA0.2 mg/L, IBA 0.3 mg/L, KT 2 mg/L and IBA 0.1 mg/L+KT 2 mg/L,6-BA 0.2 mg/L+IBA 0.1 mg/L+KT 3 mg/L, and the proliferation rate up to 2.30; With the shading time increasing, the content of chlorophyll a, chlorophyll b and chlorophyll a+b in the two clones seedling were gradually decreased; The total phenolic content of Y2 was the highest under shade 10d, and the total phenolic content of Y1 was the highest under shade 40d. In the same condition, the average shoot length and stem the proliferation multiple of Y2 were higher than Y1.3. The best rooting medium were 1/4MS+ NAA 0.4 mg/L+IBA 0.9 mg/L and IBA 0.5 mg/L+ABT 0.5 mg/L+6-BA 0.6 mg/L. In rooting medium, MS medium with ABT1# was good for rooting and the accumulation of chlorophyll. When ABT1# in the range of 0-1.0mg/L, the higher the concentration, the more promoting was. In 1/4MS+NAA 0.4 mg/L+IBA 0.9 mg/L, the rooting rate and the total content of chlorophyll were the highest, when the IBA concentration was lower than the concentration of 6-BA, and add the appropriate ABT, was conducive to the rooting of tissue culture seedling of Larix potaninii Batal.4. With the yellow subsoil in the matrix, the transplanting seedling was growth well. With yellow subsoil as the matrix, Bottle of Larix potaninii Batal. take root in high survival rate of preservation of transplanting seedlings, seedling height growth rate and the total content of chlorophyll was respectively 94%,10.33%,1.48 respectively, with yellow subsoil:the red cedar in bottle rooting coconut husk:Perlite=7:2:1 as the matrix, the preservation rate and the content of total chlorophyll was slightly lower than that of yellow soil, and significantly higher than the coconut husk:peat soil= 9:1.5. With the Different treatment time of IBA50mg/L+NAA25mg/L, the soaking time 5min was beneficial to the Larix potaninii Batal to take root, the rooting rate was 73%, and the soaking time 15min, was conducive to the accumulation of chlorophyll in the seedlings of Larix potaninii Batal, the content of chlorophyll was 1.68mg/g; the IBA 25mg/L was the concentration of growth regulator and was suitable for the Larix potaninii Batal micro cutting. The Larix potaninii Batal seedling sunder the treatment of micro Cuttage Rooting rate and the content of total chlorophyll total 70%,1.16; IBA 50 mg/L+NAA 25 mg/Ldoing rooting agent, yellow subsoil matrix was suitable for micro cutting; post management should not be late for seedling topdressing NAA.
