Objective: Development and characterization of microsatellite markers for themedical plant Scutellaria baicalensis. Investigate genetic diversity andpopulation genetic structure of Scutellaria baicalensis. Revealed the geneticcauses of geoherbs.Methods: We Study28wild group and22cultivation group of Scutellariabaicalensis, We development and characterization of microsatellite markersusing streptavidin-coated paramagnetic beads and study on genetic diversityand genetic structure of Scutellaria baicalensis using population genetics.Results: We isolated and characterized21microsatellite loci in Scutellariabaicalensis. These new microsatellite markers revealed polymorphism and couldbe applied to investigate genetic diversity and population genetic structureof Scutellaria baicalensis. The PCR amplification was carried out on theScutellaria baicalensis. We analysis the genetic diversity of wild andcultivated population using the theory of population genetics. The resultsshowed that both have high diversity. Wild population can be roughly dividedinto3groups, germplasm of clustering results with the distribution of sourcearea, and the cultivation group in no obvious geographical pattern of thedistribution.Conclusion:The geoherbs of producing unit is the genetic diversity center of Scutellaria baicalensis, and glacial scutellaria shelters. The superiorenvironmental conditions make this in other parts of the region of Scutellariabaicalensis in significant genetic differentiation,further influence thevariation of the quality of medicinal materials, and became the the originalregion, formed the geoherbs. Scutellaria baicalensis during the process ofcultivation, in different parts of the germplasm made frequent communicationhas led to higher heterozygosity, smaller genetic differentiation, the originof cultivated Scutellaria baicalensis is more center.
