Turpan black sheep commonly known as "toksun black sheep".Turpan black sheep meat quality isexcellent,is worth Promoting new mutton sheeP breeds,the research on turPan black sheep on moleculargenetics is rare. This study consists of turPan black sheeP as the research object of meat Performancerelated MSTN/Smad signaling Pathways functional genes to determine the amount of the expression ofmRNA level.This article main research content is as follows:1.by real-time fluorescent quantitative PCR technology RT-PCR analysis of turPan black sheep MSTN、Smad2, Smad3, Smad4,TGFBR1and TGFBR2gene0to6months age of black sheep and0month and6months of age turPan bicePs femoris muscle in his black sheeP,tricePs,half tendons,half membrane muscle,back muscle,the longest tail fat and other6kinds of different Parts of the skeletal muscle andliver,lung,sPleen,intestine,heart,kidney and so on6kinds of internal organization mRNA exPression levelswere analyzed,and the tyPe of MSTN/Smad signaling Pathway gene exPression characteristics andexPression Pattern of space and time.2. conventional methods histological methods muscle growth index determination of0to6months(number of muscle fibers and muscle fiber size) and meat Performance index (lean meat rate,body weight,the same flesh and blood ratio,net meat,eye muscle area),the analysis of the turpan black sheep musclegrowth and develoPment regularity and its differences with each other,and to exPlore MSTN/Smadsignaling Pathways and their muscle growth and meat Performance relationship.The results showed that:1)0month after gene correction made in different parts of the organization,including TGFBR1geneexpression in lung tissue without the gene and other genes in bicePs and triceps,half tendons,halfmembrane muscle,back muscle,the longest tail fat,liver,lung,spleen,intestine,heart,kidney and so on areexpressed in the different organizations.MSTN/Smad signaling Pathway genes are expressed in differenttissues,to have influence on the growth and development organizations.2)0to6month of leg muscle and fat tail MSTN/Smad signaling Pathway genes in leg muscle and fattissues are expressed,different months for MSTN/Smad signaling Pathway genes in different growth stagesof turPan black sheep leg muscle and fat tail of the exPress no increased with the increase of months hasbeen the trend of increase or decrease.3)Through the MSTN,Smad2,Smad3,Smad4,TGFBR1and TGFBR2gene with0~6month turpan blacksheeP in the muscle exPressed by the correlation analysis,the results show that Smad2gene expressionquantity and amount of Smad3gene exPression was significantly Positive correlation (r=0.905, P <0.01);Samd2gene expression measure MSTN and Smad4gene expression was significantly negativecorrelation (r=-0.914,P <0.05) and (r=0.957,P <0.05),MSTN and Smad2;TGFBR1,TGFBR2gene hadno significant correlation (P>0.05);Smad3gene exPression and Smad4expression was significantlyPositive correlation (r=0.848,P <0.05).4)Through MSTN,Smad2,Smad3,Smad4,TGFBR1,TGFBR2gene with body weight,body height,bodylength,chest circumference,pipe circumference,trunks,slaughter rate between correlation analysis.the resultsfound that MSTN gene expression of quantity and weight was significantly negative correlation (r=-0.993,P <0.05), and body height, body length, body weight, slaughter rate had no significant correlationbetween (P>0.05);With the very significant negative correlation between pipe circumference (r=-0.957,P <0.01);Smad2gene exPression and the slaughter rate was significantly negative correlation (r=-0.918, P <0.05),and body weight,body height,body length,chest circumference,pipe circumference,bodyweight,no significant correlation (P>0.05).Smad3gene and body weight,body height,body length,chestcircumference、 pipe circumference、 body slaughter rate have no significant correlation (P>0.05).Smad4,TGFBR1,TGFBR2exPression with body height,body length,chest circumference,bodyweight,slaughter rate have no significant correlation (P>0.05).
