| Objective:Tea saponin(TS) had a widespread biological activities,and was mainly used in chemical industry and textile industry, but less applied in medicine or animal husbandry, so in our study, toxicological experiment and pharmacology experiment are designed to provide more scientific and more comprehensive basis for the toxicology and pharmacodynamics of TS,pave the way for the more broad application of tea saponin in animal husbandry.Methods:In vitro hemolysis test, acute toxicity test, subacute test was designed to study the hemolytic activity (HD50), median lethal dose (LD50) and subacute toxicity, know more about the toxicological properties of tea saponin. Through the antibacterial test of TS combined with antimicrobial agents and the relevant tests of TS in bone marrow hematopoietic function in mice, from two aspects of antibacterial activity and promote bone marrow hematopoietic, to understand the pharmacological activity of TS.Results:1Toxicological test:From the vitro hemolysis test we know the HD50of TS is16.8ug/ml, and the LD50is bigger than5000mg/kg. And in the process of subacute toxicity test, the rat did not appear abnormal reaction, and the biochemical indexes,weight or viscera have no obvious changes, so TS may have no adverse effect on liver, kidney, etc and belong to low toxic substances. And in peripheral blood test, platelet count (PLT) and lymphocyte count (Lymph) were increased, the same results were emerged in next test.2Antimicrobial test:The MIC of TS on E. coli and Staphylococcus aureus were1.6×105ug/ml,0.8×105ug/ml respectively, and when combined with antimicrobial agents, although the concentration of TS is only reduced to0.8×10ug/ml,0.4×10ug/ml, the MIC value of gentamicin, florfenicol, enrofloxacin and ampicillin to E.coli were depressed to1/64,1/32,1/64,1/32,1/2respectively; The MIC value to staphylococcus aureus were1/32,1/8,1/4,1/8,1/2respectively.TS could significantly reduce the usage of antibiotics and have synergistic effect with antimicrobial agents.3The results in experiments about bone marrow hematopoietic function show that TS could increase the number of bone marrow cells (BMC) and the content of WBC and PLT in peripheral blood; And confirmed that TS could by increasing the content of GM-CSF, TPO, SDF in marrow cavity flushing fluid, reducing the apoptosis rate of BMC and the cell percentage in S phase to promote the hematopoiesis function of guanulocytosis and thrombocytosis in bone marrow. Conclusion:The safety evaluation of TS was first analyzed, and confirmed that TS was a low poisonous matter. And when joint with antimicrobial drugs, TS could significantly reduced the concentration of the drugs; And TS could promote the hematopoietic function of bone marrow cells. So our study provide scientific basis for the application of TS in the field of animal husbandry and medicine. |
