In this study, the branches and seeds of Haloxylon ammodendron as experimental materials, study on the disinfection of branches and its germination of axillary buds and rooting induction, effect of nutrient substance and hormone combination on the growth of Haloxylon ammodendron aseptic seedling, callus induction of the assimilating branches hypocotyl and stem tip, shoot bud induction and root induction. Discussing the assimilating branches of disinfection technology and the rapid propagation of Haloxylon ammodendron seedlings, providing technical support for Haloxylon ammodendron tissue culture breeding propagation. The results were as follows:1.The best method for the disinfection of Haloxylon ammodendron branches were(0.5%) KMnO4 15 min,(3%) NaClO10 min, antibiotics 30 min. In this way, the rate of branches survival reached 94%. After d isinfection of assimilating branches, transferring 3 ~ 4 times was the best method to get a certain number of survival assimilating branches. The rate of assimilative branches rooting induction reached 27.3% in the m edium with AgNO3 4 mg/L.2.Without casein hydrolysate(CH) and inositol(IP), the aseptic seedling leaf number of per plant and its the relative growth rate were respectively increased by 83.3%, 1 times. The treatment with 1/4MS+ sucr ose 10 g/L+ IBA 10 mg/L or AgNO3(0.5 mg/L) +IBA(5mg/L) +6-BA(1.0 mg/L) made the leaf number p er plant relative growth rate largest. Seedlings for 40 d in the MS+6-BA 0.6 mg/L+ AgNO3 4.0 mg/L, block buds number was most.3.MS culture medium was best for callus induction of the assimilating branches hypocotyl and stem tip.The callus induction rate of stem tip was better than the assimilating branches hypocotyl. MS + 2,4-D 1.0 mg/L+6-BA 0.2 mg/L was the best formula for induction callus of Haloxylon ammodendron aseptic seedlin g. MS+AgNO3 4.0 mg/L+NAA0.2 mg/L+GA3 0.4 mg/L+6-BA 1.0 mg/L was the best for induction of mult iple shoots. The effect on root induction of aseptic seedling assimilating branches was the best with IBA co ncentration in the range of 0.2~0.8 mg/L. The root induction rate from high to low was the bottom(hypocot yl), the middle(epicotyl), the upper(stem tip).
