Malus asiatica,a genus of Malus of the Rosaceae family, is a small deciduous tree, with beautiful fruit and flowers in the United States. Malus asiatica, as an endangered tree species, contains a variety of vitamins and minerals. At present, the study on the tissue culture technique of the Malus asiatica is not sufficient and the main propagation technique of Malus asiatica is grafting, which has a low propagation coefficient. To develop the propagation technique of Malus asiatica, this paper focus on the study of tissue culture technique of Malus asiatica and wish to find a good way of improving the quality and quantity of Malus asiatica propagation. This study took tissue culture technique and orthogonal design. Ann bonus stems were selected as explants to establish explants sterile system, and then go through bud induction and proliferation to take root and achieve rapid propagation purposes at last. The main results were as follows:1 The best way to sterilize bonus stems for tissue culture is: Hydroponics explants 75% alcohol 20s; explants were collected in the field with 75% alcohol 20 s + 0.1% mercuric chloride 7.5 min.2 Lai’an bonuses hydroponic explants in February showed leaf yellowing in bud induction process. Sand treatment until March then did not have this problem. Considering the sterile buds start rate and survival rate, March and May were Lai’an bonuses tissue culture’s best time for explants colleting.3 The effect of disinfecting on half-woody stems was slightly better than the effect of disinfecting on tender stem segments. And the effect of disinfecting on these two stems were better than on woody stems.4 Best solution for reducing browning rate is inoculating explants continuously transferred to fresh medium, once a day,followed by adding activated carbon to the medium and low temperature processing. Dark treatmentcould decrease browning rate to a certain extent.5 Different lignification explants start to bud induction effect: semi woody stems> tender stems> woody stems. Semi-woody stems sprouting fastest, just 6-11 days, and bud growth robust.6 Orthogonal design, found to affect the culture of the buds of the largest, followed by 6-BA, IAA minimal impact, the optimum medium for WPM + 6-BA 2.0 mg/L + IAA 0.4 mg/L, the induction rate up to 73.3%.7 Orthogonal design, found that the main factors affecting the multiplication factor is 6-BA, IBA followed, and6-BA at the 0.01 level significantly different optimum proliferation medium and hormone concentration ratio of 1/2MS + 6-BA 2.0mg/L + IBA 0.15mg/L.
