Maize haploid breeding technology is a new method to quickly obtain maize inbred lines, the biggest characteristic of it is the time can be greatly shorten on breeding process of inbred lines. This experiment will combine young embryo culture a sprout technology as well as screening of haploid and double colchicine technology, by establishing the haploid doubling of transplanting system, not only the efficiency of the double haploid was improved, but also the doubling of high survival rate was ensured, meanwhile a complete system of haploid breeding process was set up. According to the principle of induced haploid parthenogenetic inducing system, 24 groups of different material combination were designed and materials with haploid were screened out, and through different chromogenic time of materials, growth status, haploid induction rate, the best combination of materials were selected to obtain haploid.For double haploid materials, the colchicine double combinations were designed, the concentration of colchicine were 0.01%(g/L), 0.02%(g/L),0.03%(g/L), 0.04%(g/L), 0.05%(g/L), 0.06%(g/L) and 0.07%(g/L), the processing time were 24 h and 36 h. After observing the root tip chromosomes and supervising the plant survival rate as well as the doubling efficiency, the effect of root tip chromosomes double haploidwas apparent, transplanting experiment plant survival rate exceeded 90%,ultimately 0.06%(g/L) of colchicine was certified as the best concentration for doubling, furthermore, the plants grow optimally.
