The population structure, leaf and steam biomass, and quality of Acanthopanax senticosus were systematic researched, in order to reveal the population growth and development of A. senticosus, the effect of different sample area and different years on content of syringin from roots and stems were investigated, which can provide scientific basis for the syringin accumulated mechanism of A. senticosus. And the results were as following.1. At the five plots, the population biomass and density were the biggest, which were 75.03 g/m2 and 2.85 plants/m2 in the Yu yang plot, respectively. The population density and biomass only were 1.27 plants/m2 and 37.22g/m2 in the Yu yin plot; the individuals plant grew better, but the population density and biomass were the lowest, which were 23.62 g/m2 and 0.35 plants/m2 in the Xin Kailing plot, respectively. The results showed that the population density and canopy density had great effect on individual plant biomass of A. senticosus. The cultivation of the A. senticosus should be reasonable density.2. Based on the stem and leaf component simulation modeling found that the R2 of determination coefficients of the Xin xing, Xin Kailing, Yu yang and Yu yin plots with good power function relationship were 0.8366, 0.7383, 0.8555 and 0.7985, respectively.3. High Pressure Liquid Chromatography(HPLC) detection method of syringin from A. senticosus was established. HPLC conditions: Bartlett Hypersil ODS2(4.6 mm × 250 mm, 5 μm) column, detection wavelength was 265 nm, the column temperature was 25 ℃, the injection volume 5μL, methanol and water as the mobile phase ratio 20:80 and a flow rate 1.0 m L / min. The retention time of syringin was 11.8 min. The optimum extraction of syringin were studied based on the ultrasonic assisted extraction and microwave assisted extraction method by the single factor and orthogonal experiments. The optimum conditions base on ultrasonic assisted extraction were that extraction frequency was 70 MHz, extraction time was 40 min, solid-liquid ratio was 1:15; the optimum conditions based on microwave-assisted extraction were that the extraction power was 300 W, extraction time was 15 min, solid-liquid ratio was 1:10 and extraction temperature was 90 ℃. And the two different extraction methods rate under optimal conditions were compared, we found that the extraction rate based on microwave-assisted extraction method was higher.4. At the five plots, syringin contents from the stems and roots of A. senticosus tended to increase with the increasing age in the different age and plots. The syringin contents from stems were higher than roots except the 4 and 5 years old of A. senticosus plant. But the higher syringin contents in the roots were 4 and 5 years old A. senticosus plant, and then it tended to be stable with the increasing age. The syringin contents in stems and roots in Yuyin plot were higher than other plot, but the syringin yield was low which affected by high canopy density. The syringin yield was high at Yuyang plot with 50% canopy density. Therefore, the 50% canopy density habitat should be choosed in bionic cultivation.5. The population structure belonged to growth form, the population density, biomass and syringin yield in Yuyang plot were higher than other plots, In order to determine the specific influenced factors of the accumulation of population biomass and the secondary metabolism product of the A. senticosus syringin, the A. senticosus population structure of Yuyang plot should be further studied.
