The antioxidant capacity and Gene the characteristic of expression of transferring and effects of vitamin C on them were studied by the methods of developmental biology, histology, enzymology and molecular biology during early development of Carassius auratus gibelio. Following results and conclusions were achieved from these thesis:1. The experiment conducted to study effects of vitamin C on early development of Puan silve Crucian Carp(Carassius auratus gibelio). The membrane time, hatching ratio, survival rate and the characteristics of growth which sock in the vitamin C solution were studied by the method of static aqueous solution immersion. The results showed that: When the concentration of vitamin C below 30 mg/L, the membrane time of larvae were decreasing and the hatching ratio, survival rate, weight and full-length were increasing with the increase of vitamin C. When the concentration of vitamin C solution was higher than 30 mg/L. The research showed that: appropriating levels of vitamin C could promote the early development of Puan silve Crucian Carp.2. The activities of superoxide dismutase, catalase and glutathion peroxidase were studied at water temperature of 24℃ during early development of Puan silve Crucian Carp.The results showed that:(1)The activities of superoxide dismutase,catalase and glutathion peroxidase have existed in the mature eggs,and the activities were(10.213±0.458)U/mg,(8.701±0.371)U/mg and(55.485±4.896)U/mg,restectively,and belong to maternal antioxidant enzyme.(2) In the control group,the activities of superoxide dismutase and glutathion peroxidase rises gradually from mature egg to pre-hatching,while the activitiy of catalase showed “up-down-up” trend, to minimum in heart beating stage,and the activitiy was(6.175±0.532)U/mg,then the activitiy of catalase began to rise(P < 0.05), and the activity of glutathion peroxidase was showed “up-down” trend.(3) From mature egg to pre-hatching, vitamin Csolution(30mg/L) could obviously promote the activities of superoxide dismutase,catalase and glutathion peroxidase,and the activities were higher than that of control group generally, especially which significantly high than that after heartbeat stage.The research indicated that maternal antioxidative enzymes play an important role during embryo development of carassius auratus,and appropriate concentration of vitamin C in hatching water of could improve the antioxidant capacity in embryo stage of Puan silve Crucian Carp.3. The experiment was conducted to study the contents of malondialdehyde and protein carbonyl, the activities of lactic dehydrogenase and the total antioxidant capacity and effects of vitamin C on them during eary development of Puan silve Crucian Carp.The results showed that: ⑴ The contents of malondialdehyde and protein carbonyl, the activities of lactic dehydrogenase were showed“up-down”trend,and these three indexes reached hightest in mig-gastrocoel stage, while the total antioxidant capacity increased during early development of Puan silve Crucian Carp.⑵In the group of vitamin C(30 mg/L), the contents of malondialdehyde and protein carbonyl and the activities of lactic dehydrogenase were significantly lower than that of control group, while the total antioxidant capacity significantly higher than that of control group(P<0.05). The research showed that: ⑴ The oxidative damages more serious in embryonic than postembryonic of Puan silve Crucian Carp. ⑵Appropriate vitamin C solution( 30 mg/L) could improve the antioxidant capacity of embryonic and larva, and effectively reduce the oxidative damage of embryonic and larva, and improve the hatching rate and survival rate of larva.4. RT-PCR and fluorescence quantitative PCR techniques were used to detect effect vitamin C on the expression of transferring genes and its mRNA expression levels during the early development of Puan silve Crucian Carp. The results showed that:No expression of transferring in mature eggs, fertilized eggs and expanded blastocyst stage, while the expression of transferring was determined in gastrula stage,and the mRNA expression level of transferring showed “up-down-up” trend, and in the heatbeat stage to a peak, in the prehatching stage to lowest. In the group of vitamin C(30mg/L), the mRNA expression level of transferring was lower than that ofnormal group during embryonic stage, while which was higher than that of normal group during larva stage. The function of transferring was independent from the binding iron function during embryonic development, and appropriate vitamin C could make the function of transferring in immune decreased during embryonic development, while increased the function of transport iron in yolk-sac larva stage.5.The circulatory system was observed through light microscopy by morphological and histological methods during early development of Puan silve Crucian Carp, and describes the characteristic of circulatory system histological structure. The results showed that the heart of Puan silve Crucian Carp arises from mesoderm. The heart starts pumping after fertilization 28 hours, and the heart appeared as tube, which located infra-anterior to the head, the heart beats about 40-50 times per minute, and the erythrocytes start to appear.The gill filament began to appear in 3th days, blood circulation gradually increased with the development, and the big artery in the back began to form. The feeder vessel of larvae more conspicuous in 17 th days, the structural development of gill trend to complete, and the circulatory system trend to complete.
