Effects Of Supplemental Vitamin C And E In Diet Under Nitrite Stress On Immune Function Of Carassius Auratus Gibelio

The study was performed on Carassius auratus gibelio,which immune function was investigated by the nitrite stress and addition of high-dose vitamin C(500mg/kg) and vitamin E(350mg/kg) in feed under the condition of nitrite stress to Carassius auratus gibelio.In the experiment of study on effects of nitrite stress on immune function,Carassius auratus gibelio used for experiment were randomly divided into three groups.And nothing was added to the control group.However,the two treatment groups were added 0.32 mg/L and 1.28 mg/L of nitrite into water,respectively.After two weeks,effect of nitrite on the immune function of Carassius auratus gibelio was investigated.The results showed that there was no significant difference(P>0.05) about the immune organ index between two experimental groups,compared with the control group.However,we observed that it was significantly lower(P<0.01) in experimental groups than in control group for lysozyme activity in serum,the stimulation index in the head kidney and the phagocytes in the head kidney.In the experiment of study on effects of addition high-dose vitamin C(500mg/kg) and vitamin E(350mg/kg) in feed under the nitrite stress to Carassius auratus gibelio immune function,Carassius auratus gibelio used for experiment were randomly divided into four groups:control group without any nitrite in water and administration basal diets; treatment group 1 joined with 0.85mg/L nitrite in water and feed basal diets;treatment group 2 joined with 0.85mg/L nitrite in water and feed basal diets containing350mg/kg vitamin E;treatment group 3 joined with 0.85mg/L nitrite in water and feed basal diets containing 500mg/kg vitamin C in diets.The results of the tests were shown as follows:(1) Compared with the control group,the treatment 3(added high-dose vitamin C[500mg/kg]in diets),there was no significant difference about the immune organs index and acid phosphatase activity and AKP activity(P>0.05);the content of serum complement C3 and C4 were significant lower the control group(P<0.05);red blood cell count was remarkably higher than the control group(P<0.05);lysozyme activity in serum and the stimulation index in the head kidney and the phagocytes in the head kidney were significantly lower(P<0.01) in the treatment group 3 than in the control group.However, compared with the treatment group 1,we also observed that it was no significant about the immune organs index and acid phosphatase activity and AKP activity and red blood cell count and the content of serum complement C3 and C4(P>0.05);lysozyme activity in serum and the stimulation index in the head kidney and the phagocytes in the head kidney were significantly higher(P<0.01) in the treatment group 3 than in the treatment group 1.(2) Compared with the control group,the treatment group 2(added high-dose vitamin E[350mg/kg]in diets),there was no significant difference about back kidney index and acid phosphatase activity and AKP activity and the content of serum complement C3 and C4 were no significant difference(P>0.05);lysozyme activity in serum and the stimulation index in the head kidney and the phagocytes in the head kidney were significantly lower(P<0.01) in the treatment group 3 than in the control group;back kidney index and spleen index and red blood cell count was remarkably higher than the control group(P<0.05).However,compared with the treatment group 1,we also observed that it was no significant about back kidney index and acid phosphatase activity and AKP activity and red blood cell count(P>0.05);lysozyme activity in serum and the stimulation index in the head kidney and the phagocytes in the head kidney were significantly higher (P<0.01) in the treatment group 3 than in the treatment group 1;head kidney index and spleen index and the content of serum complement C3 and C4 were significantly higher (P<0.01) in the treatment group 3 than in the treatment group 1(P<0.05).(3) Compared with the treatment group 2 and the treatment group 3,there was significant lower about head kidney index and spleen index and the content of serum complement C3 and C4 in the treatment group 3 than in the treatment group 2 (P<0.05),but back kidney index,acid phosphatase activity,AKP activity,red blood cell count,lysozyme activity in serum,the stimulation index in the head kidney,the phagocytes in the head kidney were no significant difference(P>0.05).
