| B cells and T cells are the major components of lymphocytes, which are involved in the cellular immunity and humoral immunity. In this study, SPF chicken were used as the research object of interest. In order to detect the changes of T and B lymphocytes in normal developmental processes, samples of bursa, thysum, spleen, lung, and blood were collected and analyzed by flow cytometry, at the age of 1, 4, 7, 14, 21, 35, 56 and 90 days, respectively.We monitored the changes of the cell content by using flow cytometry, and the results showed that: Bu-1b cells are fewer at the age of 1 days,but at the age of 4 days the number of B cells in immune organs and peripheral blood increased rapidly.In bursa, Bu-1b cells reached growth peak at the age of 7 days.It is show that the humoral immune function enhanced in 4 to 7 days of age.In spleen, thysum, bursa of fabricius, and blood, the Ig M+ cell is the majority in the immunoglobulin of B lymphocytes. In addition, the study also demonstrated that the content of B lymphocytes becomes stable after 35 days of age.By the changes of the T cell content, the results showed that CD3+ cell content increased rapidly during the phase between 1 and 4 days, the bursal cell content decreased at 7 days, and CD3+ cell content showed little change after 35 days old which tends to be stable. There were more CD4+ cells than CD8+ cells in the blood and thymus at 14 days of age, and the same result was observed in bursa at 21 days old.The number of CD4+ cells tend to be stable at 21 days of age.To understand the immune state of chickens after incubation, we study the quantity variation of lymphocytes and their subsets in immne organs and blood of SPF chicken at normal physiological conditions by flow cytometry,which provided an important heoretical basis for the real-time immunity and disease surveillance. |
