Reticuloendotheliosis(RE), caused by reticuloendotheliosis virus(REV), is a serious immunosuppression oncosis which impaired to the poultry farming industry. At present, the REV contamination of some live vaccines, such as avian influenza vaccine and Newcastle disease vaccine, it may lead immune failure and caused heavy losses. Especi ally, when occurred mixed infection caused by REV, avian leukosis virus, infectious bursal disease virus and Marek’s disease virus, it will develop more serious disease, and caused more and more economic losses, so it is necessary to research on the pathogenic mechanism.Transforming growth factor β1(TGF-β1) is a kind of cytokine with multiple biological functions, such as, cell proliferation differentiation, tissue damage repair, bone regeneration and interstitial form, so it has been widely studied in organ fibrosis, tumor and autoimmune diseases. In this experiment, we choosed one day-old SPF chicks as the research object, polyclonal anti-TGF-β1 antibody, then the technologies of real-time fluorescence quantitative PCR and enzymelinked immunosorbent assay(IELISA) were used to respectively research the changes in the level of gene and protein of TGF-β1.We extracted the peripheral blood and immune organs of SPF chicks infected with REV. The results were as follows:(1)After 1 day-old SPF chickens infected with REV, the expression of TGF-β1mRNA in peripheral blood in 7 ~ 42 days were significantly higher than control group(p<0.01). In expression of protein, after 1 day-old SPF chickens infected with REV, the content of TGF-β1 in peripheral blood point significantly higher than control(p<0.01) of infected chicks in 14 ~ 42 days,These results indicated that early in REV infection, the expression of TGF-β1mRNA rise sharply, but the replication of the virus has affected the protein expression and secretion of the TGF-β1.(2)After 1 day-old SPF chickens infected with REV, the expression of TGF-β1m RNA in thymus, spleen and bursa of fabricius in 7 days, 7~14 days and 14 days were significantly higher than control group(p<0.01). These results indicated that after REV infection, TGF-β1 precursor compounds is activated, TGF-β1 mRNA expression has increased dramatically, the virus infected chicks immunity function is inhibitied; In the late of virus infection, thymus, spleen, TGF-β1m RNA expression gradually reduced, but the bursa of fabricius TGF-β1mRNA expression is increased.(3)After 1 day-old SPF chickens infected with REV, the content of TGF-β1 in bursa of fabricius and thymus were significantly higher than control group(p<0.01) in 7 ~ 14 days and 7 ~ 28 days. But the content of TGF-β1 in spleen was significantly higher than control group(p<0.01) in 7~21 days. These results indicated that the mechanisms of virus infected immune organ are different, it may regulate the secretion of cytokines and adapted to the life cycle of the virus.(4)Polyclonal antibody was successfully prepared.
