The grapes(V.vinifera L.) is a deciduous vine. In General, the majority of the production of grapes are mainly used in wine, dried et al. with the improvement of people’s living standards, grape has become a substantial portion of the a gricultural industrialization in China. But because of their poor quality, which was seriously restricting the widely cultivate of grapes. Therefore, Selective breeding fine grape germplasm was imminent.In this paper, selecting the robust growth of grape plantlets got from Jiayuguan Hongfeng Grape Base as test materials, such as ‘Shuanghong’, ‘Shuangyou’, ‘Beta’, ‘294’, ‘Cabernet Sauvignon’, ‘Pinot Noir’, ‘Red globe’ and ‘Cabernet France’. By using of plant tissue culture techniques and in vitro micro-grafting method, test materials were cultured under certain conditions, and by measuring related indicators, observed the influence of different rootstock and different scion on physiological and biochemical characteristics of grafted plants. Selecting grafting survival rate as the characterization factor, combined with various grafting combination indexes including the content of fructose, tannin and GA3, the activity of SOD、CAT、POD and PPO and other related indicators, the conclusions were as follow.1.The influence of different rootstock on grafted scion combinationsIn this study, Selecting the primary culture of grape plantlets ’Shuanghong’, ’ Shuangyou’, ’294’, ’Beta’ and ’Pinot Noir’ as test material, studied the impact of different rootstocks on the compatibility of grafted plants and found that the "Pinot Noir" combined with four different rootstocks, where ’ Shuanghong’, ’ Shuangyou’, ’294’, ’ Beta’ as rootstocks, ’Pinot Noir’ as scion, graft survival rate in the following order: Shuangyou> Shuanghong > Beta> 294, which showed that mountain grapes " Shuangyou" and "Pinot Noir" had the highest grafting compatibility, and the worst was "Pinot Noir" with ’294’, which may be related to the origin of species. In different healing process of grafting combinations, the physiological and biochemical indexes of superoxide dismutase(SOD), catalase(CAT), peroxidase(POD), polyphenol oxidase(PPO) activity and gibberellic etc. hormone(GA3) content, fructose, etc. were studied and analyzed.The results showed that grafting grapes in the healing process, the protective enzyme IV activity should play a protective role, the performance was that the activity of enzymes in healing part was higher than that of rootstock and scion before grafting, the order was: graft healing part> stocks> scion, the results were basically the same with other people’s study, and healing part had higher activity mainly because of the role of self-defense in healing process. The relationship between the enzymes activities in healing part and the graft survival rate showed that enzymes activities in healing part were high and graft survival rate is correspondingly high. The above results illustrated that the different rootstocks had different effects on physiological and biochemical indexes of graft combinations. This may be related to their specific grafted rootstock.2. The influence of different scion grafted scion combinationsIn this experiment, ’Beta’ as rootstocks, ’Pinot Noir’, ’Cabernet Franc’ and ’Red globe’ as scion, the healing process of grafts among different combinations were studied, through analyzing the activities of superoxide dismutase(SOD), catalase(CAT), peroxidase(POD), polyphenol oxidase(PPO) and other enzymes and gibberellin(GA3) content and other related indicators, and combining with the graft survival rate to explain the grafting compatibility strength, according to changes in the compatibility of the relevant indicators, the results showed that different scions also affect the physiology and biochemistry indexes of grafted plants, thereby affecting their survival rate. This indicated that plants were not always in the hurt state in adversity, they primarily through synergies of SOD, POD, PPO and CAT to make radical at a stable level, thus preventing free radical damage, so that aerobic organisms could survive. For the new grafted plants, the specificity of rootstock and scion, metabolism activity, organic matter content were changed, which would benefit to improving the quality and yield of the grape cultivation.3. Establishment of Integrated Assessment System on in vitro graft compatibility of GrapeIn this study, To establish integrated assessment system on graft compatibility of grape, selecting the robust growth of grape plantlets as test materials, with ‘Shuangyou’,‘Beda’ as rootstock and‘Pinot Noir’, ‘Cabernet sauvignon’,‘red globe’as scion. By using cleft grafting method, evaluated SOD、POD、CAT and PPO as well as the related indexes such as GA3, tannin and fructose content of the tube grafted seedlings after being grafted with 14、21、28、35 and 42 d, and determined the effective scope of the compatibility index of the micro-graft seedlings by combining the dynamic changes of the analysis of the grafting survival rate and the compatibility index in the growth process of post-grafting. And determined the 7 major indexes related to the evaluation of grape grafting compatibility with the method of correlation analysis and the grafting survival rate as the index of the compatibility of micro-graft grape seedlings. With the principal component analysis method, integrated the 7 indicators into the main component and determined the index weight. Besides, established the evaluation system of grape grafting compatibility by subordinate function and obtained the comprehensive evaluation index Y of each grape grafting compatibility by using subordinate function value. According to the comprehensive index Y, the compatibility order was as follow:‘Beda+red globe’> ‘shuangyou+red globe’> ‘Beda+cabernet sauvignon’> ‘Beda+Cabernet Sauvignon’>‘Shuangyou+Cabernet Sauvignon’>‘Shuangyou+Pinot Noir’> ‘Beda + Pinot noir’. This was completely consistent with the result of using the grafting survival rate as identification index. The result showed that the evaluation of grape grafting compatibility by subordinate function had certain practical significance. Therefore, it will be more accurate and scientific to comprehensively evaluate graft compatibility of grape using the method of principal component analysis in combination with subordinate function.
