| Pseudorabies is caused by pseudorabies virus(PRV) with severe sporadic or local foci and spread by contaction. As the unique PRV storage reservoir, piglets usually cause death to 100% after infection. It is an important epidemic disease, and remains a big threat to the pig industry economy.In order to further understand the PRV prevalence in our country, we used PCR to detect PRV in our country. The positive were further amplified the virus genes. The sequence was analyzed by using bioinformatics analysis software DNAStar for their homology, by using Clustal W algorithm of MEGA 5.0 software to build the evolutionary tree.The 1005 sapiens were from 185 pig farms of 21 provinces in China in 2014, and some others in 2012 and 2013 preserved in our laboratory. Also we analyzed their protein homology and built the evolutionary tree based its amino acid sequence. The positive pig farms were 22.2%, and the positive rate was 12.4%. We got 22 g B, out of them, 20 encods 915 aa, and they had inserts in 280-282 and 93-93, and deletions in 74-78 aa, their similarity was 96.8% to 97.0%, nucleotide similarity was 97.9% to 98.0%, and in different branches with Bartha strains. 16 g C genes encoding 488 aa with 7aa insertion in 62-70 aa, their similarity was 93.1%, nucleotide similarity was 95.8% to 95.8%, and in different branches with Bartha. 15 g E genes encoding 580 aa, and had inserts in 1488-1490 aa compared with Ea, their similarities was 99.1% to 99.1%, nucleotide similarity was 99.6% to 99.8%, and in different branches with the prevelence Ea, La, Fa, Min-A. 10 TK genes encoding 321 aa, their similarity was 99.1% to 99.4% compared to Bartha, nucleotide similarity was 99.6% to 99.7%, and in different branches with the prevalence Bartha, La and Min-A.Results showed that PRV infection is still widely spred in our country. The amino acid variation is bigger, the g B and g C variation is bigger than the virulence genes g E and TK. Whether these variation will affect its infection is still unknown and needs further study. |
