Doritaenopsis,which is hybridized by Phalaenopsis and Doritis,belongs to the Phalaenopsis, a category of orchid. Doritaenopsis has relatively high ornamental and economic value due to its luxuriant design, influent color, graceful shape and long flowering time. This research put seven-months healthy butterfly orchid clone seedlings called ‘Tinny Tender’ under different lighting time and autonomous condition,and cloned the genes which are related to the flowering process under the photoperiod and autonomous way. This study also quantitatively studied the samples of various flowering period and some genes, and determined the mutual relation of different genes expression during the process of butterfly orchid flowering in time and space.The main outcomes of this study is as follows:1.Under low temperature condition( 22 in daytim℃ e and 18 at night) ℃,different light length treatments have noticeable influence on the date of Phalaenopsis extracting pedicel. The result showed in 35 th day the long day 16/8 h(day/night) group and short day 8/16 h(day/night) group was in vegetative growth phase while the contrast group 14/10 h(day/night) began to extract pedicels; in 48 th day more than a half control group was extracting pedicel and the short day group had extracted pedicels, while the long day group started extracting pedicels until the 63 rd day. In addition, in the way of autonomous induction,Doritaenopsis with no low temperature treatment started to extract pedicels until the 132 day.2.The colonized genes of photoperiod and autonomous way include SOC1,FY,FCA,OsFY, OsFCA,FT,FVE,FPA,LD,FLK,DRM1,PRP36,FLD,FLC,CO,PHYB,LHY,LFY,ELF3, FKF1,ZTL,LUX,Hd6,GI,TOC1,Ehd1,CRY2,FWA,AP1,etc. Homologous comparison indicated that colonized genes have high homologies with Arethusa.3.The result of the quantitative study of photoperiod way showed that under different light length, the flowering suppressor genes decreased with the process of plant growth, while flowering promoting genes turned out a increasing trend.Certain differences are found for suppressor genes and promoting genes of three treatments, which led to the difference of smoking infarction period, growth stage of leaves, the length of stalks and the number of bud during the flowering process. It also found genes such as FPA,LD,FCA,FY,DRM1,FVE,FLK and FLD had different expression pattern in autonomous way.In addition, the analysis showed the quantity of FLC firstly increases and later it turns out a decreasing trend, while the expression of SOC1 gene showed a low level at first, after that it enhances gradually,which match the butterfly orchid flowering period.4.The related experiments of Phalaenopsis in photoperiod way indicated the biomass status, the content of ZR and the soluble saccharide change of control group are higher than the other two groups, while the content of GA and IAA are lower than the other two groups.
