Pueraria is an excellent cultivated plants with many applications containing soil and water conservation, biomass and medicinal and edible. To meet the needs of new indigenous species Pueraria lobata (wild) Ohwi ’Dabashan’ in expand cultivation, this test take the stems with buds as explants, used completely random design and orthogonal design combine with analysis of variance and comparaisons multiples, the establinment of sterile cultures, primary culture, multiplication of buds and formation of root were researched. Moreover, The tissue culture seedlings were treated with low phosphorus, in order to investigate effects of phosphorus stress on growth and physiological characteristics of Pueraria lobata (wild) Ohwi ’Dabashan’. The results showed that:(1) Potted the root tuber of excellent Pueraria lobata (wild) Ohwi ’Dabashan’ in field and placed in the greenhouse, disinfected the materials with 0.1%carbendazim solution at primary growth in Spring and this can contribute to the establinment of sterile cultures.(2) The explants were first rinsed 30 min by running water followed 30 min immersed by 0.1%(W/V) detergent solution, then rinsed 2h by running water after brushed the dust on the surface. Rapid immersed with 75% alcohol for 20s plus 0.1%HgCl2 for 10 min and then rinsed 5 times with sterile distilled water can control the contamination rate of 14.55% and the highest survival rate was 67.34%.(3) The best medium for the primary culture was 1/2MS+6-BA0.5mg/L+IBA0.3mg/L with the induction rate up to 92.2%.(4) MS+6-BA1.0 mg/L+NAA0.01 mg/L+IBA 0.05 mg/L was the most suitable medium for buds multiplication from primary culture while value ratio reaches 5.0.(5) Take the buds higher than 2 cm from multiplication culture for rooting, the optimum for rooting which rooting rate achieved 96.00%was 1/2MS+NAA0.01 mg/L+ IBA 0.1 mg/L.(6)Under low phosphorus stress, the biomass and heights ofPueraria lobata (wild) Ohwi ’Dabashan’ were both decreased as well as the contents of soluble protein while the root top ratio, the contents of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD),MDA and soluble sugar were increased. Take fuzzy membership function to evaluate the resistance ability under low phosphorus synthetically, mild aphosphorosis can enhance the resistance ability under low phosphorus of Pueraria lobata (wild) Ohwi ’Dabashan’,. moderate aphosphorosis and heavy aphosphorosis can enhance the resistance ability under low phosphorus in primary, and with prolonged phosphorus deficiency the resistance ability under low phosphorus would reduced and damaged on the growth. It is showed that the growth of seedlings were inhibited in a certain extent under low phosphorus stress and manifested some morphological and physiological adaptation,In conclusion, the establishment of rapid propagation technique for Pueraria lobata (wild) Ohwi ’Dabashan’ by tissue culture and the effect of seedlings under low phosphorus stress on on growth and physiological characteristics in the research, this can provide a based technology for expanded production and theoretical reference for phosphorus fertilizer management in production cultivation.
