Lentinula edodes(Berk.) Pegler was a kind of wood rot fungi which can be used as both edible and medicinal, it’s the world’s second largest edible fungus after the production of Agaricus bisporus. Main agricultural products in Henan Province was a major agricultural, it’s output of major agricultural products ranks the forefront of the country, such as grain, cotton and oil crop, which was using agricultural wastes as raw material development of L.edodes provides adequate protection of raw materials. At present, L.edodes cultivation were mostly used in the off-season cultivation mode, the used of L.edodes varieties should to be further developed and studied in the Henan province area. In this paper, the Henan provincearea five strains of L.edodes as the object of study, analysis and comparison of nutritional characteristics, extracellular enzyme activity changes and agronony characters of the five varieties, starting from the angle of enzymatic explore L.edodes association in the growth process of extracellular enzyme activity change and agronony characters, to provide theoretical basis for the selection of the optimal selection of L.edodes varieties, biological transformation, breeding and so on. The results were as follows:(1) Five kinds of L.edodes ’s use of carbon sources were not the same, the optimum carbon of 931-9 and Lingxian-1 were sucrose, the optimum carbon of 9608 and L808 were dextrose, Nanshan-1was maltose. Five L.edodes varieties were consistent on the use of nitrogen sources, organic nitrogen was better than inorganic nitrogen, beef extract was the best comprehensive performance. When C/N in the range of 10/1-30/1, five kinds of L.edodes mycelium have good growth, the ratio of 20/1 to the best. When the N concentration in the medium was 0.04%-0.08%, the growth rate of L.edodes was the fastest, and the mycelium growth potential was best in the high concentration range.(2) The L.edodes mycelium under three kinds of optimum liquid culture medium,it was found that four kinds of extracellular enzyme activity change presented certain regularity. Enzyme activity peak appeared successively, Amylase activity peak appeared firs, followed by CMCase, Half cellulose and Laccase. This shows that the use of nutrients in the process of the growth of L.edodes mycelium had a certain order, starch was used earlier than cellulose and lignin. The extracellular enzyme activity of different strains in the medium of three of the most suitable culture were different, but did not follow the regularity.(3) The variation regularity of different extracellular enzymes activity of the five kinds of L.edodes remain the same. Four kinds of enzyme activity changes in the presence of a complementary relationship, indicating that the L.edodes ’s used of nutrition in the order was starch and lignose in vegetative growth stage was the first to be used, cellulose and hemicellulose in the reproductive growth stage was degraded.(4)There was a certain difference between the agronony characters of five strains of L.edodes, The train cycle of Nanshan-1, L808, 931-9 were the shortest, the yield of Nanshan-1 was the highest, L808’s shap was the best, 9608 and Lingxian-1’s context were pachyntic and tight.(5)The correlation analysis of agronony characters and the extracellular enzyme activity showed that in vegetative growth stage, amylase, laccase, CMCase’s activity had great relevance with the time of L.edodes into the reproductive growthstage.The greater the enzyme activity, the more fully the mycelium on the decomposition of the culture, the more beneficial to L.edodes early fruiting. Laccase was the key enzyme, the higher the activity, L.edodes turnning color sooner, the sooner the fruiting. In reproductive growth stage, extracellular enzymes activity had little relevance with the time of L.edodes into the reproductive growthstage. In vegetative growth stage, Half cellulosea ctivity was bigger and more conducive to the formation of shiitake production, its activity also and pileus diameter and stipe also certain correlation with fruiting period sooner or later are not very relevant.
