Monokaryons of 4 cultivated strains and 6 wild strains of Lendnula edodes were obtained via protoplast monokaryotization technology. Crosses were made among 10 monokaryons which had been chosen one from the ten dikaryotic strains in allpossible combinations , C102, and 36 dikaryotic strains were gained by mono-mono mating among these ten monokaryotic strains.All dikaryotic strains were inoculated on the media using starch and glucose as main carbon sources respectively, and different concentrate 2-deoxy-glucose (2DG) was added. It was carried out to compare their colony growth rate, average diameter of hypha, mycelia biomass and glucose consumption on the media using glucose as main carbon source. Their resistances to 2-deoxy-glucose were measured, and the effects of different concentrate 2DG to those characters above identified.The result shows that among 46 strains used in this study, eleven are resistant and nineteen are strong resistant to 2DG using starch as main source; while all parental strains are resistant to 2DG and all hybrid strains are strong resistant to 2DG using glucose as main source.A randomized block test with 4 replication was made for approaching the relation between production and 2DG resistance. Comprehensively analyzing the results, the conclusion was that most strains which form fruiting bodies easily, fruit early and yield highly are resistant to 2DG on the media in which starch is main carbon source.
