Medicinal plant genetic resources bank is the conservation facilities that preserves medicinal plant germplasm resources, and low temperature genetic resources bank is the best way to preserve the medicinal plant germplasm resources. Evaluation and security save of germplasm resources are the basic work of genetic resources bank. This study based on the quality evaluation and preservation of Cyathula officinalis Kuan seeds for provide the technical specification and reference base to run the National traditional chinese medicine genetic resources bank.1. Producing ecological environment survey of Cyathula officinalis Kuan Undertaked field investigations in main producing areas, and compared the ecological factors of different producing areas.The results showed that Cyathula officinalis Kuan likes cool and moist climate, enjoys ample sunshine and rainfall, and mainly distributes in humid climate that the annual average temperature is in 11℃~19℃, the annual precipitation is more than 1000mm and the elevation is in 1200m-2500m.2. Quality evaluation of Cyathula officinalis Kuan seeds Measuring the clonerity, thousand seed weight, water content, germination rate and seed vigor of the 18 batches Cyathula officinalis Kuan seeds from different producing areas.And the measured results of the indicators are analyzed through K class center clustering analysis method. The results showed that exist significant difference among germination rate and seed vigor of 18 batches seeds, at the same time, according to the K class center cluster, got the center value of the four index, and then based on the center value to divide Cyathula officinalis Kuan seeds quality standard into four grades. And the germination rate, the seed vigor, and the thousand seed weight are given priority to determine the quality of Cyathula officinalis Kuan seeds.Analyzed the correlation between the seeds quality and ecological factors, and the results showed that the ecological factors and the connection degree of seed quality is very close with annual relative humidity, altitude, annual average sunshine, annual average temperature, annual rainfall, annual evaporation, they are in the process of growth of Cyathula officinalis Kuan, bigger influence on the quality of Cyathula officinalis Kuan seeds.3. Study on preservation of Cyathula officinalis Kuan seeds Cyathula officinalis Kuan seeds as experimental samples, this study based on suitable drying method, the optimum moisture content and appropriate preservation methods are systematically study:Different drying method and drying temperature drying seeds, get the suitable drying method of Cyathula officinalis Kuan seeds that is mechanical drying before save and the drying temperature is 50 ℃. Prepared seeds of different water gradient and artificial accelerated aging the seeds of different water gradient,research quality of different water gradient seed before and after aging, the result shows that the optimum water content of Cyathula officinalis Kuan seeds storage preservation is 5.5%~6.0%.By artificial accelerated aging treatment, research the Cyathula officinalis Kuan seeds chromosome aberration phenomenon, explore the decay in the process of Cyathula officinalis Kuan seeds preservation. Different preservation methods save Cyathula officinalis Kuan seeds, measure the seed quality after 2 months, result shows that the Cyathula officinalis Kuan seeds appropriate preservation methods is low temperature preservation, save the follow-up seed quality in the process of monitoring will be done by the team members to further. The principle of these methods and can be run in the national Chinese medicine germplasm bank (chengdu) provide technical specification and reference.
