Diarrhea in piglets is conditioned disease, which restricts the development of pig industry. Preparation of traditional Chinese medicine has a long history, broad prospects, no residue, environmental protection and other advantages, has become a focus of attention, and can effectively replace antibiotics in livestock and poultry breeding industry has become an important measure of sustainable development. This paper studied the effect of the treatment of Colibacillus diarrhea in piglets and preliminary application in piglets.1. Isolation and identification of pathogenic Escherichia coliFecal collected from the six-nine breeding pig farm of Chongqing, which was separated to obtain the pathogenic Escherichia coli with eosin methylene blue Medium after managing, and bacterial colonies, morphological, biochemical identification and molecular biology identification, at the same time through animal pathogenicity test to screen out the pathogenic Escherichia coli. The test results showed that pathogenic Escherichia coli of eight strains were isolated from pig farms collected fecal sample-s.16S rDNA sequence of OJ-1 that was selected compared with Escherichia coli, Enterobacter spp., Salmonella, Shigella and other related bacteria of the genus 16S rDNA sequences of representative strains, the similarity of OJ-1 strains and ETEC is 100%.2. Effect of traditional Chinese medicine preparation for the treatment of Escherichia coli diarrhea in pigletsThe present study was performed to investigate the preventative effect of Chinese herbal preparation on diarrhea that caused OJ-1 strains of Escherichia coli that isolate-d return to the piglets. Fifty piglets (21 d old, and initial BW of 4.41 ±0.01 kg) were assigned to 5 groups (control group; low dose; Medium dose; high dose and antibiotic group). Then using lianmei dysentery traditional Chinese medicine preparation that was composed of five dried Chinese herbs, including Fuctus Mume (Wumei), Coptis chinensis Franch. (Huanglian), Cortex Fraxini (Qinpi), Fructus Chebulae (Hezi) and Barberry Root (Sankezhen), Which were all purchased from Chongqing Dongtian Pharmaceutical Co. Ltd (human, China). It is used in the treatment of diarrhea in piglets, and observation of curative effect of the traditional Chinese medicine preparation for diarrhea disease. The test results, the tradition-nal Chinese medicine has a certain effect on Escherichia coli diarrhea in piglets, check the statistics Log-rank and Breslow check method are 4.87 and 4.00, and their p values were 0.30 and 0.41, which indicated that the traditional Chinese medicine for increase of survival rate of piglets had no significantly difference (p> 0.05). From the survival curve, the control group of the curve is steeper, which has a high mortality; traditional Chinese medicine preparation in dose curve is relatively flat, with a longer survival period.3. Preliminary study on Application of traditional Chinese medicine preparationIn order to study the effect of compound Chinese medicine preparations on growth performance, diarrhea rate, blood biochemistry and the effects of intestinal flora. Seventy-five piglets, were equally divided between male and female,21 d old, were randomly assigned to one of the 5 treatment groups, with 3 replicates in each group. Respectively, control group, low dose group of Chinese herbal, middle-dose group, high-dose group and antibiotic group. The results show that, (1) The average daily feed intake of each group compared the difference was not significant (p>0.05); the level of ADG between multiple alignment, the difference between the middle dose group and the control group were significant (p<0.05);each group was no significant difference for the feed weight ratio (p>0.05)(2)add 0.75% of compound Chinese herb diarrhea piglets and the rate of control group and low dose group were significantly higher than those of (p<0.05), but no significant difference with the antibiotic group and high dose group (p>0.05); (3)for RBC of blood add 0.5% of the traditional Chinese herb had extremely significant difference compared with the control group (p<0.01), the content of ALT, AST and TG of each group in serum has no significant (p>0.05).(4)in middle dose group, T3 content of piglets compared with the control group significantly in serum (p<0.05),and each group haven no significant difference for T4 content; (5) The contents of IgG and IgA in serum of piglets can be significantly increased in middle dose group;(6)The number of Escherichia coli in the cecum, the antibiotic group compared with the control group had significant difference (p<0.05); the number of lactic acid bacteria, the middle dose group and the antibiotic group compared with the control group, there was significant difference (p<0.05); the number of bifidobacteria, there was no significant difference between groups (p> 0.05). The number of Escherichia coli in piglets in the ileum, antibiotic group was significantly lower than that in control group and high dose group (p<0.05; the number of Lactobacillus and Bifidobacterium, between groups no significant difference (p>0.05). The use of PCR-DGGE technology to analyze the Lian-mei dysentery preparation of traditional Chinese medicine on weaning piglets intestinal flora diversity effect, results show that the piglet feed, even may traditional Chinese medicine and antibiotics, can increase the diversity of intestinal microflora.
