| This paper studies the taxis mechanism of Bactrocera dorsalis (Hendel)which attracted by ME after irradiation on pupae of B. dorsalis. The results showed that after irradiation, the taxis of infertility male reduced. Different irradiation dose and irradiation in different pupa age of B. dorsalis resulted different decrease of taxis. And after irradiation, the expression pattern of odorant binding protein (OBP) in the antennae of the male changed.1. With 100 Gy irradiation dose of 137Cs, irradiated-2 d male pupae of genetic sexing strain, we had recorded the taxis to ME after 1 d emergence of infertility male B. dorsalis, but the taxis was weaker than normal males. While after 7 d emergence of wild B. dorsalis, the taxis to ME showed up and the taxis was significantly weaker than infertility male. After feeding ME to infertility male, the day age for taxis to ME delay to 3 d and in the 13 d, the taxis was as well as infertility male.2. With different irradiation dose (90 Gy,100 Gy,110 Gy), irradiated different pupa age (-3 d,-2 d,-1 d) of B. dorsalis, the taxis to different concentration of ME (5 μLã€10μLã€20 μLã€40 μLã€80 μL) was recorded after 8 d emergence. The results showed that, pupa age of irradiation had significant positive correlation with the tropism of infertility male to sex lures, with the same irradiation dose, pupa age lager, the taxis to ME was more significant, while the pupa smaller, the taxis to ME was weaker; The irradiation dose had negative correlation with the tropism of infertility male to sex lures, with the same pupa age, the irradiation dose larger, the taxis to ME was weaker. Concentration of ME had positive correlation with the tropism of infertility male to sex lures., the attraction rate was increased along with the high concentration. So, if we want to get infertile male which have the similar sex lures tropism of fertile males, we should choose the lower irradiation dose and higher irradiation age of pupa.3. Using real-time fluorescent quantitative PCR technology to explored the expression pattern of odorant binding protein gene BdorOBP2, BdorOB?3 in the antennae of the male after irradiation. Results showed that, after Id emergence, BdorOBP2 and BdorOBP3 expression level significantly increased in the antennae of infertility male, and different OBP genes had different expression patterns. |
