As a precious mushroom, cordyceps militaris contains many active components, such as cordycepin, adenosine, cordycepin polysaccharide, cordycepic and ergosterol,and was proved to have the same medicine value as aweto. In the present paper, we focused on the optimization of extraction technics of these major active components and culturing conditions, with aim to improve the quality and utilization of cordyceps militaris. The main achievements in the paper are as follows:1. The effect of light intensity and temperature on the content of cordycepin in cordyceps militaris, were examined. Using the cordyceps militaris growing at normal culture condition for 40 days as the test object, the best combination was achieved through orthogonal test:the light intensity is 4000 lx, the temperature 25℃, treated for 9 days. The content of cordycepin in the treated strains increased from 0.445 mg/g to 2.187 mg/g.2. The effects of light intensity and temperature on the content of 5 major active components in cordyceps militaris were further investigated. The data showed that, under the comprehensively optimized condition as followed:light intensity is 2000-3000 lx, the temperature 20-25℃, the content of all 5 major active components in cordyceps militaris reached a high level.3. The extraction technics of cordycepin has been comprehensively optimized. Based on the effects of these factors on the yield of cordyceps, such as extract mode, extraction times, extraction time, liquid to solid ratio, pH, different concentration of ethanol, different concentration of carbinol and the drying temperature. The best extraction technique was obtained as follows by orthogonal test:using the distilled water as extractant, the liquid to solid ratio is 100, and extract twice, and extract time for 30 min, and the drying temperature 50 ℃, consequently the extraction rate of cordycepin is about 2.714‰.4. The comprehensive extraction of the 5 major active components, cordycepin, adenosine, cordycepin polysaccharide, cordycepic, and ergosterol was investigated. Comparing the effects of various extraction methods on the yield of the 5 major active components, the best extraction technology is:after extraction with distilled water for one time,60% ethanol was used, extraction time is 60-80 min, extraction temperature 50-60 ℃, the liquid to solid ratio 60-80. The yield of cordycepin, adenosine, cordycepin polysaccharide, cordycepic, and ergosterol is 2.184 mg/g,0.922 mg/g, 33.01 mg/g,208.05 mg/g,3.51 mg/g, respectively.
