Buffalo sperm freezing tolerance is poor, semen cryopreservation reduced semen quality and yield, this hindered milk buffalo large area to promote in south of China. The semen cryopreservation diluent is an important factor in semen quality of buffalo. So it’s very urgent to find out the better of buffalo semen cryopreservation dilutions. In the production practice buffalo semen cryopreserved using the same dilution of cattle. But the physical and chemical properties of buffalo and cattle sperm are very different, buffalo sperm freezing tolerance worse than cattle. Therefore, looking for the better of buffalo semen frozen dilution is very necessary on the basis of cattle frozen semen dilution. Melatonin and vitamin C are added to semen cryopreservation dilutions played a good protection. There are not yet proved vitamin D added to the semen cryopreservation diluents can improve the quality of semen,but many in vivo experiments show that the content of vitamin D and is closely related to sperm motility. What’s more, there are vitamin D receptors on sperm. Inferred that added this to semen cryopreservation dilutions can improve the quality of frozen semen. The experiment will add melatonin, vitamin D and vitamin C to buffalo semen cryopreservation dilutions, detecting sperm motility, sperm motility and sperm abnormality. Study its effect on sperm cryopreservation of buffalo. 一、Effects of melatonin, vitamin D and vitamin C added to the semen cryopreservation diluents of buffalo1. Adding melatonin to buffalo semen cryopreservation dilutions(10-3,10-4,10-5mol/L) can improve buffalo semen quality. Concentration of 10-3 and 10-5mol/L melatonin motility rate was significantly higher(P <0.05).Wherein the Concentration of 10-5mol/L melatonin(58.0%) is best, compared to the control(46.7%) increased the motility rate 24.28%. 2. Adding vitamin D to buffalo semen cryopreservation dilutions(10-3,10-4,10-5mol/L) can improve buffalo semen quality. The Concentration of 10-4mol/L vitamin D(56.4%) is best, compared to the control(46.7%) increased the motility rate 20.85%. 3. Adding vitamin C buffalo to semen cryopreservation dilutions(4.00,0.40,0.04g/L) can improve buffalo semen quality. Concentration of 4.00 and 0.40mol/L melatonin motility rate was significantly higher(P <0.05). Wherein the Concentration of 0.40mol/L melatonin(59.7%) is best, compared to the control(46.7%) increased the motility rate 28.01%.二、Effects of melatonin, vitamin D and vitamin C combined with each other4. Buffalo semen cryopreservation diluent added melatonin and vitamin D can improve the quality of buffalo semen, but the effect is bad compare with melatonin or vitamin D. 5. Adding melatonin and vitamin C in buffalo semen cryopreservation dilutions can improve the quality of buffalo semen. What’s more, significantly better than the single addition of melatonin or vitamin D. The effect 10-4mol/L melatonin combined with 0.40g/L vitamin C added is best, VCL,VSL, VAP also significantly higher(P<0.05), highest sperm motility(63.2%) in all treatment groups, compared to the control(46.7%) increased 35.47%. 6. Buffalo semen cryopreservation diluent added vitamin D and vitamin C can improve the quality of buffalo semen, but the effect is bad compare with vitamin D or vitamin C.
