Ambient temperature storage of boar semen is a key technique of artificial insemination(AI), the preservation effect is directly related to sperm quality and fertilization ability. However, sperm can be affected by the reactive oxygen-induced damage during the preservation, which may results in damaging to sperm membrane, acrosome and motility.In contrast, boar sperm are sensitive to cooling, the rapid cooling could cause cold shock, which results in an irreversible loss of motility and disruption of membraneand acrosome. Therefore, protective agents added for semen is very important. The objective of this study was to indicate the effect of resveratrolon Duroc spermatozoa. The results, detected by fluorescence microscopy, flow cytometry and scanning electron microscopy, show below:1. During the ambient temperature storage, we found the sperm motility, membrane integrity, acrosome intactandmitochondrial potentials in treatment group(50μM resveratrol)elevated significantly compared to control groups(P<0.05), the intracellular reactive oxygen species(ROS) showed a remarkable decrease in treatment group(P<0.05), superoxide dismutase(SOD) activity of sperm in treatment group and control group had no significant difference between treatment group and control group(P>0.05), it shows that resveratrol could protect the sperm from oxidative stress.2. After rapid cooling, the sperm treated with 150μM resveratrolexhibited preferable indexes in motility, membrane integrity, acrosome intact,mitochondrial potentials and total antioxidant capacity(P<0.05). Furthermore, the 150μM resveratrol treated group produced less intracellular ROSand malonaldehyde(P<0.05).Meanwhile, we have resolved the ultrastructure of spermatozoa under the scanning electron microscopy, found that morphological structure of sperm head and neck in treatment group is more integrity than control group. It reveals the protective effect of resveratrol on boar sperm against cold shock.In conclusion, this research indicated that boar sperm supplemented with resveratrol could scavenge intracellular ROS, protect the spermatozoa against the reactive oxygen-induced damage, inhibit the decrease of sperm quality during the ambient temperature storage and cold shock treatment, the optimum concentration of resveratrol is 50μM and 150μM, respectively.
