Opisthopappus taihangensismum,growth in the cliffs,steep slope,the aperture of bare rock,due to habitat and steep,population reproduction and hum an over-exploitation in the endangered state, is the key protection plant in henan province.Its use value mainly include medical and ornamental.Using tissue culture technology can reduce the production cost,improving the efficiency of m icropropagation.And you can get rid of the influence of natural climate change,overcome the Opisthopappus taihangensismum harsh natural conditions in the wild habitat cond itions,and the bottom of the sm all amount of seed,germination rate,breeding,with th e best breeding efficiency production seedling product quickly,for nature reserve Opisthopappus taihangensismum species protection,city garden plant ornam ental and Opisthopappus taihangensismum medicinal plant resources de velopment and utilization to provide some useful technology, as an ornamental and medicinal resources and so on also has the very good market and industrialization prospect.Therefore, this test from seed disinfection sterile sy stem to study the in vitro rapid propagation technology,then to induce ro oting and dom estication transplanting,a series of studies provide a b asis for the sys tem in the Opisthopappus taihangensismum tissue culture technology,specific results are as follows:(1) The seed disinfectionStudy the influence factors of the Opisthopappus taihangensismum seed disinfection,the conclusion is as follows:first,the Opisthopappus taihangensismum seed soaking to sterile water for four hours,then super clean workbench with 75% alcohol rinse 15 s quickly,then use 0.1% m ercuric chloride(plus two drops of twain- 20) 8 min,finally rinse 3-5 times with sterile water.(2) Multiple shoot clumpsThe seeds grow no vaccine induced m ultiple shoot clumps,through orthogonal test the best formula for multiple shoot clumps value-added: MS + 6- BA1.0 mg/L + NAA0.1 m g/L + sugar 30 g/L+10 g/L agar strip, pH 5.8, value-added ratio can reach 3.78.(3) Take root seedlingTo grow to more than 4 c m somaclone moved to rooting medium,take root seedling obtained by orthogonal test the best formula for MS + NAA0.2mg/L + 30 mg/L sugar +10 g/L agar strip, pH 5.8, gr ow stout and strong, about 5 cm long, 4 weeks will take roo t somaclone transplanted into MS culture medium.
