Abstract/Summary:
Tomato(Lycopersicon esculentum Miller) is one of the important vegetable crops in the world, and it suffers from various diseases due to continuous cultivation. In this research, we have made a seven kinds of diseases 9 resistance genetest in 56 tomato materials through molecular marker technology, which is year-cultivated.Combining the field observations traits andselecting elite inbred lines formulated crosses by spring and autumn crosses test, we have filtered the three good combinations in spring and autumn three good combinations, and constructed autumn three excellent combination of ISSR DNA fingerprinting maps according to botanical characters,fruit quality hybrid combination of principal component analysis, and the main component of the composite score,. The test results for the post-screening suitable for multi-resistant tomato grown in our region to provide technical support. The main results are as follows:1.Among 126 tomato materials, the number of including tobacco Mosaic virus disease resistance genes Tm-2α materials is 33. These tomato materials have 4 homozygous yellowing leaf virus resistance genes Ty-1 and 7 hybrid. The number of these tomato materials, including hybrid Ty-2 resistance gene, is only 1. Due to Ty-3 of resistance gene, the number of the homozygous genes is 5 and the hybrid is 7. In terms of fungal diseases, include leaf mildew resistance gene Cf-9 materials, have taken over the most, reaching 50. The number of late blight resistance genes homozygous Ph-3 isl, while the hybrid is 6. The number of these checked materials, which includes Fusarium Wilt resistance genes 1-2, is 32, the number of Verticillium wilt resistance genes homozygousVe-2 isl, while the hybrid is 4. These tomato materials have 16 units, which contain resistant to root knot nematode resistance genes of Mi-1.2. Through statistical observation in 27 spring-crosses unites’33 traits, different traits are varied in different crosses.Based on principal component analysis, the 32 traits were divided into five main components,.the accumulating the main ingredient in the composite score, all ranked cross have filtered 3 elite crosses— C27, C12, C22. Observating 22 crosses unites in 35 autumn traits, we find a cross of traits between the coefficient of variation is large,. Through principal component analysis, the 35 characters are divided into six main components, the composite score based on the main ingredient, we have flitered Q15, Q1, Q43 anti-yellow leaf curl virus disease is a good cross of field.3. Selecting from 100 ISSR primers,we found three obviously polymorphic primers--UBC-11, UBC-16, UBC-34. Combining Q15, Q4, Q1, it seems that their parents were amplified with 17 clear and stable bands, and 12 bands were polymorphic and the percentage of polymorphic bands is 70.59%.Using a single primer UBC-11, UBC-16 and UBC-34 can distinguish combination Q4, Q1 and its parents. At the same time,using a single primer UBC-34 and two primers UBC-11, UBC-16 can make a difference between Q15 and its parents.
