Aronia melanocarpa Elliot is adaptability and strongly resistant to cold, which is suitable for planting in most areas of China. Fruit is rich in a lot of biologically active substances and anthocyanins, can maintain our healthy heart, and prevent cancer, and prevent the hypertension, and can slow down human aging, so that the body maintain a healthy body. At present, the progress in Aronia melanocarpa Elliot is rapid, but induction in terms of polyploid has not been reported. Therefore, this study use chemical method, combining tissue culture technology, select different materials, get on the polyploid induction, explore the induction process of Aronia melanocarpa Elliot ploidy regulation mechanisms, as well as polyploid identification, provide for our country to cultivate varieties of germplasm resources, to expand the planting area, to meet the market demand has important economic significance and value. The results showed that:1. Comparison of different materials on polyploid induction, when different materials was founded polyploid induction, using Aronia melanocarpa Elliot callus as inducing material than shoot tip, leaf; comparing the way of using colchicine polyploid induction of colchicine method was applied to the high survival rate colchicine in immersion method, plant has low damage.2. Using Aronia melanocarpa Elliot stem tip as material induced polyploidy, the regenerated plants were preliminary identification. Results showed that:mutant plants stomatal density was significantly lower than the CK plants, guard cell length and width were significantly higher than the CK plants, the number of chloroplasts is 12 to 20 plants, plants can be assumed to polyploid variation.3. When soaking concentration was 1500 mg · L-1, the processing time was 36 d, the resulting regenerated plants from leaves in the chloroplast mutation rate was the highest, reached 33.3%. When it was 200 mg · L-1, the processing time was 14 d, the highest induction rate reached 35.6%.4. Colchicine immersion induced polyploidy total was 47, of which 7 is mixoploid, tetraploid cells was accounted for 12.5%; 60 facilities into law induce polyploidy, which has 13 of the mixoploid chimeras, tetraploid cells was up to 23.5%; the other found three polyploid, the rate of induced can be 11.1%.5. Following the polyploid plant after three generations, its stem diameter greater than diploid plants, the difference between the length of the section was not significant, leaf length and width were significantly higher than diploid plants.
