A sample survey was conducted in buffaloes came from backyard households and large scale milk buffalo farms in BeiLiu and QinZhou Districts to investigate the health status of milk buffalo in these two districts, and to explore the clinical meanings of buffalo’s blood biochemical indexes such as liver function in the process of health examination. Furthermore, the differences of biochemical figures between plasma and serum were also identified in this study, as well as the figure changes of plasma in longtime conservation. The study was conducted with four parts. In the first part of the experiment, blood biochemical parameters of liver function of milk buffaloes were measured in BeiLiu City. With 46 milk buffaloes in 19 farms,25 came from dairy farms and the left 21 came from backyard households. It was found that the levels of serum aspartate aminotransferase (AST), serum alanine aminotransferase (ALT) and albumin (ALB) in milk buffaloes from dairy farms were significant higher, and total protein (TP) was extremely significant higher than that of milk buffaloes from backyard households. With higher activities of ALT, AST and GGT in the detected buffaloes, the activity abnormal rate of each was 23.9%,21.7% and 47.8%, and the levels of TP and ALB in buffaloes from dairy farms were significant higher than the normal reference values. Then the preliminary conclusion is made that the detected milk buffaloes are in good wealth overall and the liver function of backyard households’ buffaloes are superior to that of dairy farms’. Abnormal check indicators such as liver enzyme and protein may be related to the vaccine 1 month ago. The buffaloes’ blood biochemical figures indicating liver and kidney functions were detected in QinBei District in experiment two. As the objects of study,25 buffaloes were selected from 12 farms in QinBei District. The results were found that the activity abnormal rates of ALT and AST were 92% and 100% respectively, and the rate of higher levels of TP, GLOB and Urea were 36%,88% and 16%. It was also found that buffaloes in dairy farm were seldom dewormed and mainly fed in grazing manner during the survey. The conclusion is that the abnormal liver function may be related to the vaccination that some buffaloes had 40 days ago, and may be related to the seldom scientific deworming in buffaloes from dairy farms during long-term grazing; but its specific reason remains further study. Part three were the analysis of some biochemical indexes in plasma and the investigation of culturing situation in large-scale milk buffalo farms.96 milk buffaloes were selected to detect the function of liver and kidney and field trip and survey in farm was also conducted in this study. It was found that the levels of ALT, AST and AST/ALT were all increased to 47.97±18.8(U/L),165.02 ±42.89(U/L) and 3.75±1.75(U/L). With average levels of ALI、AST、GGT、TP and GLOB all significant higher than the normal reference values, their abnormal rates(on the high side) were 84.4%、96.9%、71.9%、82.3% and 68.8% respectively. It is found that the abnormal situations above may be related to the inoculation 40 days before detection and long-time’s feeding of wine by-products after investigation. The conclusion of above three experiments is that some potential health threatening of buffalo can be found by detecting liver function, and pathogens of buffaloes’liver function in every experiment are remained further study. Experiment four was about the differences of biochemical indexes between serum and plasma, as well as the changes of plasma after long-term storage. With 25 buffaloes selected in the experiment, the levels of TP, ALB, GLOB and UA in serum were significant lower than that of plasma (P<0.05) when compared the biochemical indexes of serum with that of plasma, and the other indexes had no significant changes. The detection values of AST had no significant differences between samples cryopreserved at -80℃ for two months and the samples collected within three days (P> 0.05), and the levels of ALT, GGT, TP, ALB and GLOB were no significant differences between samples cryopreserved at -80℃ for 1 month and 2 months(P> 0.05). The conclusion is that during the detection of TP, ALB, GLOB and UA in plasma, corresponding reference range values are recommended to establish to differ from that of serum. The levels of ALT, AST, GGT, TP, ALB and GLOB in plasma are in stable states after one month’s storage at -80℃ condition, suggesting that if blood samples can be quick cryopreserved and their indicators still have clinical significance.
