This study aimed to get the suitable addition of yeast cultureby in vitro and in vivo tests,and evaluate the effect of two kinds of yeast culture : XP, YC1 on rumen fermentation and nutrient flow and digestion in the gastrointestinal tract of sheep. Nine sheep with an average weight(35±2.67)kg, fitted with ruminal duodenum and ileum cannulas for test. The paper is divides into three parts as follows:First, The suitable addition of yeast culture was to get under the basal diet with C/R ratio of 3:7 by in vitro and in vivo tests and comparing YC with DiamondV XP. By using two factors cross group design,The test was divided into two phases:The first part was in vitro experiment. The test was divided into five groups( I,II,III,IV,V) based on the concentration of YC(0%、1%、1.5%、2% and t 2% DiamondV XP). Each group had three parallel.The second part was in vivo experiment. The test consisted of three groups inclouding:the control group and the top two leading concentration of yeast culture in vitro test and each group had three parallel.The concentration of pH,NH3-N,and VFA were measured for each sample of in vitro and in vivo in the 3 times.The in vitro results showed that on the whole tend, the effect of 1.5%、2% yeast culture on rumen fermentation was similar to DiamondV XP. Significantly decreased NH3-N concentration, increased acetic acid, propionic acid, butyric acid concentration(P<0.05).And the in vivo results showed that 1.5% group can significantly increased TVFA concentration, decreased NH3-N concentration(P<0.05).1.5% yeast culture was the suitable addition.Second,This study aims to evaluate the effect of two kinds of yeast culture(YC) : XP, YC1 on rumen fermentation of sheep. Treatments were 1) The control group; 2)feed 1.5%YC1; 3)feed 2% XP. The results showed that the effect of group I、group II yeast culture were Significantly increased pH(P<0.05), There was no significant effect of pH in group I and group II. The concentrations of NH3-N in group II were significantly lower than in group I(P<0.05), The concentration of NH3-N in group III was significantly lower than in group I(P<0.01)and group II(P<0.05). group II and III can increased BCP concentration(P>0.05), acetic acid, propionic acid concentration(P<0.01). The results suggest that YC can stabilize rumen pH value and increased BCP(P>0.05)、TVFA concentration(P<0.01), decreased NH3-N concentration(P<0.01).Finally,This study aims to evaluate the effect of two kinds of yeast culture(YC) : XP, YC1 on nutrient flow and digestion in the gastrointestinal tract of sheep. Treatments were 1) The control group; 2)feed 1.5%YC1; 3)feed 2% XP. Both kinds of YC reduces the rumen liquid flow rate, rumen liquid volume and the residence time and rectum digesta DM, OM flows(P<0.05), XP reduces the rectum ADF flow(P<0.05). Both kinds of YC increase ruminal digesta MN and duodenal MN flows and DM, OM, ADF and NDF digestibilities in total tract(P<0.05). YC increases CP digestibility in small intestine(P<0.05). The results suggest that YC is by improving the digestibility of nutrients in the small intestine to improve the total tract digestibility.
