The experiment was conducted to study the effect of Flammulina velutipes stembase(FVS) on the growth performance and lipid metabolism of broilers.AA broilers of serum biochemical indexes related to lipid metabolism, abdominal fat, hormine sensitive lipase activity in abdominal fat andthe contents of fat and cholesterol in liver, muscles of broilers,FAS,ACC,HMGR,SREBP-1 and ATGL key gene m RNA expression level in liver were determined to comprehensive measure chicken lipid synthesis and decomposition level of FVS on broilers.Then the verification that whether the AMPK signaling pathway is activated promoted fat mobilization.The objective of this study was to explore a reasonable development and utilization of industrial waste materials FVS,and to provide theoretical and experimental basis, the production of Green pollution-free feed additives.A total of 270 one-day-old Arbor Acres broilers were randomly divided into 5 groups with 3 replicates of 18 chickens each. The 5 treatment groups were: the control group(basal diet), the antibiotic group(basal diet+ 5mg/kg Flavomycin), and 3 levels of FVS group(basal diets containing 2%, 4%, 6%FVS),the experiment lasted for 42 d. The results showed that:1) Effects of FVS on growth performance of broilers:On 1-21 days,each dosage FVS group and antibiotic group could significantly increase the average daily gain,average day intake(P<0.05) and reduce the feed conversion rat(P>0.05).On 22-42 days, 6% FVS group could significantly increase the average daily gain.Each dosage FVS group could significantly increase average day intake and reduce the feed conversion rat(P<0.05).The results showed that the addition of FVS could improve the growth performance of broilers.2) Effects of FVS on the serum biochemical indices and fat deposition of broilers: Dietary supplement with 2% and 6% of FVS significantly decreased triglyceride(TG) in the serum(P<0.05). Each dosage FVS significantly reduced total cholesterol(TC) in the serum(P<0.05)and significantly increase the content of leptin(LEP) in the serum. but Each dosage FVShad no significantly influence on the high density lipoprotein(HDL-C) and low density lipoprotein(LDL-C)(P>0.05). Different treatment group significantly increased the body weight of the broilers(P<0.05) and significantly reduced lipid content in liver, thigh muscle, and breast muscle(P<0.05), but had no significant effect on the excreta of broiler(P>0.05).The abdominal fat deposition was significantly reduced in the broilers fed with 4% and 6% FVS group(P<0.05), and the activity of HSL was significantly enhanced in the broilers fed with 2%, and 6% FVS group(P<0.05).On 1-21 days,FVS group have no significantly influence on the content of FFA, while on 22-42 days 2% FVS group could significantly increase the content of FFA.The results of this study suggested that FVS was able to reduce serum lipid and lipid deposition of broilers.3) Effects of FVS on cholesterol metabolism of broilers:in 21 days old,6%FVS group could significantly decrease the content of cholesterol in breast and significantly increase the content of bile acid in excreta of broilers. Compare to the control group,each dosage FVShave no significantly decrease the cholesterol content of thigh,hepatic,excreta of broilers. Whereas,Compare to others group, the antibiotic group could significantly increase the content of cholesterol. In 42 days old,each dosage FVS group and antibiotic groupcould significantly decrease the cholesterol content of hepatic.Compare to the control group,each dosage FVS group and antibiotic grouphad onsignificantly effect on the cholesterol and bile acids in the chest, leg and stool of the broilers. The results of this study suggested that FVS could decrease the content of cholesterol in muscle and hepatic of broilers.4) In 21 and 42 days old,6%FVS group could significantly increase AMPK protein levels of phosphorylation but 2% and 4% FVS have no significantly influence on AMPK protein levels of phosphorylation.Each dosage FVS could significantly decrease them RNA expression level of FAS,ACC,HMGR and SREBP-1and significantly increase the m RNA expression levels of ATGL. The results of this study suggested that:FVS could inhibit the expression of fat synthesis enzymesgenes and promote lipolysis-related gene expression through the AMPK signaling pathway activated by AMPK protein phosphorylation.So AMPK may involved in the regulation of lipid metabolism in broilers.In conclusion,FVS, as the feed additives, could improve the growth performance of broilers,decrease the content of fat and cholesterol in serum and muscle. AMPK signaling pathway activated by AMPK protein phosphorylation that FVS made could promote lipolysis through inhibiting the synthesis of lipids in the liver of broiler.
