With the carrot pomace, bagasse and bran as raw materials, the flammulinavelutipes mycelia fermentation conditions and the mycelia polysaccharide extractingconditions was analysised. The anti-tumor and antioxidant activity of thepolysaccharide were studied. The results were as follows:1. With carrot pomace and bran as raw materials, the optimum condition ofliquid fermentation was researched. The optimum liquid fermentation condition wasthat the ratio of carrot pomace to bran content was3.5∶1, the initial pH was7.0, theliquid medium volume was120mL/250mL, the cultural time was7days. Themycelia maximum dry weight achieved2.044g/100mL. With bagasse and bran asraw materials, the medium of solid fermentation was that the ratio of bagasse to brancontent was4∶1.2. The condition of extraction flammulina velutipes mycelia polysaccharides wasstudied. The optimum water extracting condition was that the solid-liquid ratio wasl∶40, the extracting temperature was85℃, the extracting time was4.5h, the initialpH was6.0, the volume of ethanol as three times as polysaccharides liquid was usedto precipitated, extracting two times. The maxium of extraction rate achieved2.19%.3. Three removing protein methods of Sevag method, trichloroacetic acid andpolyamide method were contrasted. Using polyamide method to eliminate protein inthe crude polysaccharide, protein removal rate was higher, polysaccharide loss ratewas less. Bsides, it was non-toxic, simple operating, reusable, and could keep thebiological activity of polysaccharide. So it was the optimum choice. With8%polyamide, protein removal rate was up to66.13%. In addition, the polyamide had theeffect of removing pigment.4. The effect of FV-1(water extraction from flammulina velutipes mycelia),FV-2(crude polysaccharide of flammulina velutipes mycelia)and FV-3(crudepolysaccharide without protein of flammulina velutipes mycelia) on Bel-7402cellproliferation was sdutied by MTT colorimetric method. The three components had some suppressed effect and presented doseeffect relationship. The order of antitumoreffect was as follows: FV-2>FV-3>FV-1. When the concentration of FV-2was640μg/mL, the inhibition rate of Bel-7402cells was up to45%. With clearance rate ofDPPH· and·OH as index, antioxidant effect of FV-1, FV-2and FV-3were studied.The three components had some antioxidant effect and presented doseeffectrelationship. The sequence of radical scavenging activity was as follows:FV-3>FV-2>FV-1. When the concentration of FV-3was2mg/mL, DPPH· clearancerate was65.85%,·OH clearance rate was71.24%.
