Study on the optimum of flask culture technology, the concentrations of metabolic product of submerged fermention product, and the antioxidant and antibacterial activity of fermentation product, of medicinal mushroom Phaeoporus obliquus (Ach. ex Pers.) J. Schrot, were studied in this paper.Our team further optimized the flask culture technology of Phaeoporus obliquus with both the one-factor-at-one time and the orthogonal matrix methods on the basis of former work in the library, the result show the optimal fermented condition was:tincubation time of seed liquid 15 days for mycelia growth, The optimum medium for the biomass accumulation was made up of 55g/1 of glucoses,2.5g/1 of soy peptones,0.25g/1 of MgSO4-7H2O,0.46g/L of KH2PO4,1.5 g/L of K2HPO4,0.25 g/L of MnSO4·H2O; the optimum medium for the exopolysaccharides production was made up of 70g/1 of glucoses,2.5g/1 of soy peptones,0.25g/1 of MgSO47H2O,1.38g/L of KH2PO4, 2.0g/L of K2HPO4,0.5 g/L of MnSO4·H2O.The ultrasonic extraction method of total flavonoids from Phaeoporus obliquus and its extraction process were studied. The effects of ethanol concentration, the material-liquid ratio, extraction time, and extraction temperature on the extraction of total polyphenols and total flavonoids from Phaeoporus obliquus were studied by using ultrasonic extraction method.The best extraction conditions for polyphenols and flavonoids extraction were confirmed by using orthogonal experiment and single factor test. The best extraction conditions of polyphenols were as follows:40% ethanol as the extractant, material-liquid ratio of 1:25, extracting at 60℃for 45 min; while the extrac condition of flavonoids were 40% ethanol as the extractant, material-liquid ratio of 1:25, extracting at 60℃for 60 min.Under the optimized conditions, the concentration of polyphenols, flavonoids and triterpenes in mycelium were 4.57mg/g,1.25 mg/g and 62.35 mg/g respectively; while the result in sclerotium were 8.94 mg/g,3.72 mg/gå'Œ53.12 mg/g respectively.The nutritional condition of accumulating triterpenes in Phaeoporus obliquus was studied.The result show that P. obliquus accumulating triterpenes while used starch as carbon source and yeast extract as nitrogen source. pH had no effect on synthesis of triterpenes. In salt stress condition, the concentration of triterpenes in mycelium of P. obliquus increased.The antioxidant activity of submerged fermentation products and sclerotium of Phaeoporus obliquus were investigated and compared. The effective components of submerged fermentation products and sclerotium of Phaeoporus obliquus was extracted with petroleum ether, ethyl acetate, butanol and water as the solvents resp. And antioxidant activities of the 10 extra fractions were evaluated by hydroxyl radical scavenging assay and 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging assay. The result showed that the extra fractions possessed with hydroxyl radical scavenging activities and DPPH radical scavenging activities, the ethyl acetate fractions and the butanol fractions showed better antioxidant activity than other fractions. The antioxidant activity in extras of sclerotium were higher than in extras of submerged fermentation products.The antibacterial activity of submerged fermentation products and sclerotium of Phaeoporus obliquus were investigated and compared. The effective components of submerged fermentation products and sclerotium of Phaeoporus obliquus were extracted with petroleum ether,ethyl acetate,butanol and water as the solvents resp. Perforated method was adopted to detect the antibacterial activity and the minimum inhibitory concentration(MIC) of 10 extrafractions. The result showed that the extra of Phaeoporus obliquus had inhibition on aureus staphylococcus and bacillus subtilis and the ethyl acetate fractions and butanol fractions showed evident antibacterial effect,but all of the water fractions had no antibacterial effect. The antibacterial activity of the extra of mycelium was similar to the extra of Sclerotium.
