Background and ObjectiveOrthotopic liver transplantation(OLT) in the rat is a common model for the study of immune rejection and Ischemia-reperfusion injury . In 1973 , lee successfully established the model of liver transplantation in rats by suture in anastomosis of portal Vein and hepatic inferior vena cava ., but the model is so difficult that it did not get promotion very well . In 1983 , kamada applied cuff technique to OLT and greatly simplify the process of the model . However , kamada did not reconstruct the hepatic artery. Some scholars argued that without the reconstruction of the hepatic artery in OLT , the risk of complications such as biliary ischemia will Increase , Without arterialization in OLT also will lead to adverse effect in liver histology and intrahepatic biliary system structure . Therefore, the arterial reconstruction in OLT begun to attract scholars’attention . Although in the past two decades , many scholars published new methods and technology about arterialization in OLT , the reconstruction of hepatic artery is still difficult to complete . It require not only the sophisticated equipment and operating under the microscope , but also a good partner . This experiment is to explore a more convenient technical methods of arterial reconstruction.Materials and MethodsForty male SD rats weighing 320g—400g were used as donors and recipient . Based on kamada’s cuff technique , a modified“sleeve”anastomosis between the celiac trunk of donors and common hepatic artery of recipients were performed to reconstruct hepatic artery blood flow .ResultsSucceeded in using the modified sleeve anastomosis technique to establish the arterialization of orthotopic liver transplants in rats ,a success rate of 85%,75% for the week survival . The average operative time : Donor(58±6)min,in vitro repair time for the liver(14±2)min , receptors(50±7)min , anhepatic phase(19±3)min , modified sleeve anastomosis(3±1)min . 20 cases of formal experiment , in which three cases of death, and two of them died of respiratory arrest caused by excessive anesthesia, one of them died of hemorrhagic shock. 17 rats survived in the two cases, one case of death was hepatic artery anastomosis louxue ; another was portal vein stenosis. After one month, 15 rats were sacrificed not found in artery embolization except one case .ConclusionsThe modified sleeve anastomosis is repeatable,simple and high patency rate. It is a new method of arterial reconstruction in rat liver and should be promoted .
