This paper took macroporous adsorption resin adsorption rate and desorptionrate as the indexes to screen out the best resin for enriching the flavonoids fromElaeagnus angustifolia Blossoms. The optimal process conditions wereinvestigated through single factor test.DPPH· method was used to compare the freeradical scavenging-capacity of every part from Elaeagnus angustifoliablossoms.We gave priority to the most active part to isolate. Monomer compoundswere isolated from the most active component.Scavenging abilities of monomercompounds from every part were also studied. In view of Tilirosides from theethyl acetate part in large quantities, we have studied its antioxidant effect on theD-galactose aging mice.1. DA201resin showed good adsorption and separation property for theflavonoids of Elaeagnus angustifolia blossoms. The detailed process conditionswere as follows: the ratio of diameter to height of resin column was1:8, sampleloading rate was0.5mL·min-1, sample concentration was20mg·mL-1, pH wasadjusted to4, with elution rate of0.5mL·min-1, the largest sample load was4.5BV, eluent concentration was80%and the dosage was12BV. DA201type resinshowed good purification effect for the flavonoids of Elaeagnus angustifoliablossoms in the defined process conditions with a yield of85.23%. The purity oftotal flavonoids achieved14.95%, which was3times of that by coarse extraction (4.901%).2. Scavenging ability of70%alcohol extract and its extraction part fromElaeagnus angustifolia blossoms was as following: ethanol extract> water solublepart≈n-butanol extract> ethyl acetate extract> petroleum ether extract. Then-butanol part was further separated and the most active component was F,fromwhich compound1is isolated. From the other components we found compounds2and3. From petroleum ether part were3kinds of compounds and from Ethylacetate part were4kinds of compounds.They were L-quebrachitol(1),2-phenylethyl-β-D-glucopyranosid(e2),uridine (3),β-sitosterol (4and8),28-norepinephrine lupeol(5),daucosterol (6), Tiliroside (7),oleanic acid(9).Compound5.Compound10is now being elucidated in.3. The purposes of experiment were to observe the effects of Tilirosides onlearning and memory functions、 index of serum, brain index and the index ofimmune organs in D-galactose aging mice.Results displayed only a small dose ofTilirosides(1mg/ml)can significantly improve the activity of serum SOD,but themiddle and high dose-group had no significant changes, which indicated theeffective dose of Tilirosides may be at low concentrations. In order to prove thatthe experimental results were associated with improper doses related, smallerdoses will be conducted in the subsequent experiments.
